The cold is one of the most universal stressor in northern animal,which is an important factor to restrict northern swine farming development leading to the direct and indirect economic loss.Sus scrofa have more adaptive capacity than swine in the disadvantage environment such as cold stress and other bad conditions.HSP70 and GR are the most important substances in stressed organism,they can produce a marked effect in the process of cytoprotection,cellular damagement,immunoregulation and Inflammatory reaction.so it is important to reveal the different of antireversion force between swine and Sus scrofa through studies on the change rules of molecules.witch played the most important roles in stress.This experiment apply fluorescence quota PCR technology to observe heat shock protein 70,glucocorticoid receptor mRNA expression change in the blood leukomonocytes from swine and Sus crofa with different cold stress intensity(0℃,-5℃,-10℃),to approach realations between the HSP70,GR mRNA expression level and the cold stimulation intensity,to provide the theory and the experiment on reveal the different of antireversion force between swine and Susscrofa.According to the Sus scrofa HSP70,GR gene sequence,primers and probes were designed,and quantitative research HSP70,GR mRNA in swine and Sus scrofa blood leukomonocytes after cold stress using real-time fluorescence quantitative PCR.The result showed,with temperature decreasing,HSP70 mRNA was significantly increased (P<0.05) while GR mRNA was significantly decreased(P<0.05),and Sus scrofa has more HSP70 mRNA and GR mRNA than swine at the same temperature.We conclude that there has obviously difference between the expression of HSP70,GR mRNA in swine and Sus scrofa at the same temperature.Therefore,this effect has been explained as a reason of why Sus scrofa has better adaptivity than swine in cold conditions.
