For knowing the effects of transport on antioxidative ability in piglets, weselected 18 Erhualian and 12 Pietrain piglets at about 20Kg body weight asexpriment animals. There were 9 Erhualian and 6 Pietrain piglets in experimentgroup and the others were arranged in control group. The piglets in experimentgroup were transported for 2 h. Then all of piglets were killed after anaesthesia withPentobarbital Sodium.The samples of livers and hearts were collected and keeped at-20℃or -70℃. ROS in the samples were measured with the method offluorescence luminescence, MDA was measured with the method of TBA, T-AOC,CAT and GSH-PX were measured with chemistry colorimrtry, SOD andCu-ZnSOD were measured with the method of hydroxylamine.The results were as follows: The activities of T-AOC, CAT in the Pietrain andErhualian's livers were lower than those of the control group(P<0.05). Theconcentration of LPO in the Pietrain and Erhualian's livers was significantlyincreased after transport stress (P<0.05). The levels of ROS and the activities ofSOD, Cu-znSOD and GSH-PX in the Pietrain and Erhualian's livers were nosignificant difference after transport stress (P>0.05). There are no significantdifference between the two breeds.The levels of ROS, MDA and the activities SOD, Cu-znSOD and CAT in thePietrain and Erhualian's hearts were no significant difference after transport stress(P>0.05). The activity of T-AOC in the Pietrain's heart was significantly increasedafter transport stress (P<0.05). The activity of T-AOC in the Erhualian's hearts wasno significant difference after transport stress (P<0.05). There are no significantdifferences between the two breeds.The expressions of Cu-ZnSOD gene in the hearts and livers of experimentgroup were lower than the control group, but there were no significant differencebetween them (P>0.05).The above results indicated that the degree of lipid peroxide was strengthened,while the antioxidant capacity was weakened, the ability of resisting free radical of Erhualian was better than that of Pietrain in heart and the liver after transportation.The decrease in expression Cu-ZnSOD mRNA induced the depression of antioxidantcapacity in the tissues after transport stress.
