Effects Of Casein Hydrolysate And Insulin On The Intestinal Growth And Development Of Newborn IUGR Piglets

Abstract/Summary:
The experiment compared the difference of weight and length of small intestine,the tissue morphologic of small intestine,the content of DNA,RNA and protein in small intestine mucosa,the activity of lactase,maltase and alkline phosphotase in small intestine mucosa and the concentration of total protein,urea nitrogen,alkline phosphotase,glucose,insulin and IGF-I in plasma among newborn IUGR piglets bottle-fed for 3 days with cow-milk and cow milk supplemented with casein hydrolysate or insulin in order to investigate the effects of casein hydrolysate and insulin on the growth and development of small intestine in IUGR piglets.The first experiment: Sutai piglets was used as the test animal. Fifteen IUGR piglets were selected from five litters and allotted randomly to three groups: newborn group N, casein group C and casein hydrolysate group CH. The intestinal length of piglets in group CH was significantly higher than that in group C (P<0.05). The villus height/crypt depth of distal jejunum and primal ileum in group CH was significantly higher than that in group C (P<0.05). The protein and DNA contents in small intestinal mocosa in group CH were significantly higher than those in group C (P<0.05). The DNA concentration in small intestinal mocosa in group CH was higher than that in group C (P=0.056). The activity and realtive activity of maltase in small intestinal mocosa in group CH were significantly higher than those in group C and group N (P<0.05 or P<0.01). The activity of AKP in small intestinal mocosa in group CH were significantly higher than than in group C (P<0.05). These results suggested that casein hydrolysate can stimulate the hyperplasia of epithelium cell and increase the activity of maltase and AKP in small intestine mucosa.The second experiment: Sutai piglets were used as the test animal. fifty IUGR piglets were selected from five liters and divided into newborn group N, milk group M and insulin group I. The difference of intestinal weight,mucosa and non-mucosa weight among three groups do not get the significant level of statistics (P>0.05). The villus height of primal and distal jejunum in group I was higher than that in group M (P<0.05 and P=0.065). The villus width of distal jejunum in group I was significantly higher than that in group M (P<0.05). The contents of DNA,RNA and protein in small intestinal mocosa in group I were significantly higher than those in group M (P<0.05). The activity of lactase,maltase and AKP in small intestinal mocosa in group I were significantly higher than those in group M (P<0.05). The results show that oral insulin can improve the growth of the intestinal villous and development of intestinal mucosa in newborn IUGR piglets.
