In this study, we investigated the extraction of DNA from young leaf and fruit, attempting to establish a optimal system of high-quality genomic DNA isolation. This will provide a technical basis for the conservation, indentification, estimation and utilization of plant genetic resourse, also giving a widespread application of plant cultivar protection, marker-assisted selection in breeding and many molecular downstream researches.This study optimized the main factors influencing DNA extraction in order to establish a optimal system of DNA extraction. The experiment has designed four processing, the three processes used the CTAB method, processes 1 is DNA extraction of different variety, processes 2, different CTAB contranctions, processes 3 DNA extraction of hyacinth beans leaf and fruit, fourth processes the contrast of the CTAB and the SDS methods. In processes 1, the DNA examination was quite good of the variety 3; In the different CTAB density, 2% CTAB extraction buffer is better; The leaf and fruit DNA extraction, the leaf was better than the fruit; the contrast of the CTAB and the SDS methods, DNA which the CTAB method to extracted are purer, pollute few. The optimi DNA extraction result is the variety 3 leaf and 2% CTAB extraction buffer DNA quality good, the purity is high.
