Biological Characteristics And The Genetic Sequence Detection Analysis Of S1 Gene Of Avian Nephritis IBV KIBVXJ Isolated From Xinjiang

In this study, the virulence of the pathogenic KIBVXJ strain (including pathogenicity tests of chick-embryo) was examined. By renal cell pathogenicity tests of chick-embryo and the pathogenicity tests of chicken),. The results showed that KIBVXJ caused the typical respiratory apparatus pathological variation and had very strong virulence for chick-embryo renal cell, and revoked typical systema respiratorium symptom for experimental chicken. The reseach of the immunogenicity showed that KIBVXJ strain has favourable immunogenicity, and may stimulate animals to create specific immune antibodies.It was obtaind a total length of S1gene for KIBVXJ strain about 1700 bp by RT-PCR amplification. Sequenced and analyzed its S1 gene genetic evolution showed that KIBVXJ S1 gene sequence have more amino acids replacement, insertion and deletion phenomenon; the cleavage site of the S1 gene was HRRRR sequences, and have characteristics of the local pandemic strain in China. Genetic evolution analysis showed that the phylogenetic relationship was very far with the major domestic and foreign vaccine strain, and had the closely relationship with the domestic A2 strains, LX4 and QXIBV strain, it could be a new variant strain.With the Newcastle disease -bigem of the infectious bronchitis oil-inactivated vaccine the experimental chicken were inoculated, and sera were collected the regularly, and the ND antibody was detected by hemagglutination inhibition test (HI), and IB antibody was detected with indirect ELISA test. The results showed that the vaccine can stimulate the animals to create favourable ND and IB specific antibodies. The Newcastle disease -bigem of the infectious bronchitis oil-inactivated vaccine has a good the prevention immune effect for ND and IB.
