Pepper(Capsicum annuum L.)is one of the most important vegetable crops in the word,the output in our country occupy first in the word..Pepper was cosmopolitan vegetable and condiment,nutrition was enrichment.Our country attach importance to pepper breeding,since twenty eigthy's,we have breed about 100 pepper cultivars,the primary cultivars have renovated 3-4 times,these were very important to increase output and change character.The conventional breeding was limited intraspecific,and genes were centralized in few germ plasm,genetic Basis was more and more strait and morphologic shape was more and more similar going with new cultivar,the new cultivar identify absent evidence as they don't have novelty and specificity.In recent years,with the deep research of molecular biology ceaselessly,using Molecular Marker identify cultivars has showed huge potential.Molecular Marker can identify cultivar in any growth and development period and improve efficiency.In this paper,AFLP analysis was applied to identy 12 cultivars in Capsicum frutescens L mainly planted in Hangzhou and analyse their Genetic Relationships.Main botanical traits was also observed to do clustering analysis and correlation analysis.The main results of the experiment were as fellows:(1)Correlation ananlysis of main charactors in pepper.20 main characters in pepper were analysed by bivariate method in SPSS16.0 soft'ware.The results showed plant height was positive correlation with leaf width;leafstalk,leaf length*width was positive correlation with leaf length,leaf width,leafstalk;Flesh thick was negative correlation with plant height,leaf width,average single fruit weight was negative correlation with the fiuit count of single plant;Fruit width and the fruit count of single plant were positive correlation with prophase output.(2)clustering analysis of phenotypic traits in pepper.We used data matrix of 23 phenotypic traits in pepper to clustering,genetic similarity was 0.54~0.93,average 0.75,Standard Deviation 0.09,Coefficient of variation 0.12.(3)There are much hydroxybenzene and amylase in pepper and they introduce DNA degradation in the process of DNA isolation,and the results of other people were different,so SDS method,improved CTAB methods treatment A(adding 1%2-Mercaptoethanol and 1%PVP-360 into CTAB extraction reagent)and treatment B(adding 0.2%DTT into CTAB extraction reagent)were compared.DNA extracted by SDS method had a A260/A280=1.0-1.3 detected by ultraviolet radiation absorbance and DNA drag greatly in the gelose electrophoresis;DNA extrcted by treatment had a A260/A280=1.3-1.7 detected by ultraviolet radiation absorbance and DNA drag in the gelose electrophoresis;Only DNA extracted by treatment had a A260/A280=1.75-1.95 detected by ultraviolet radiation absorbance and got a wider and bright band in the gelose electrophoresis.(4)Total 64 pairs of primer combinations which consisted of 8 EcoR I primers and 8 Mse I primers were used to screen the AFLP marker,As a result we found 10 pairs of primer combinations which bands were distinct and polymorphic.We got 96 polymorphic bands and got phylogenetic relationship tree topology by SPSS16.0 software,genetic similarity was 0.2~0.88,average 0.52,Standard Deviation 0.16,Coefficient of variation 0.31.we can distinguish 12 pepper cultivars by 15 polymorphic bands,the difficult was correlation with phylogenefic relationship.(5)The correlation analysis between genetic similarity based on AFLP markers and phenotypic traits indicated they were significantly correlated.
