Cotton disease-resistant breeding is an important aspect of genetic improvement. This study focus on mapping of QTLs conferring resistance to Fusarium Wilt in cotton and analysis of Fusarium oxysporum f. sp. Vasinfectum. First of all, the 32 gathered Fusarium oxysporum f. sp. Vasinfectums of Xinjiang was taken RAPD cluster analysis to establish its genetic relationship,and screened high resistance cotton varieties 98134.Then, the Gossypium.hirsutum L. 98134 and susceptible Xinhai 14(Gossypium.barbadence L. ) for parents, build 98134×Xinhai 14 F2 and F2: 3 separate groups . Construction the SSR linkage maps, and take QTLs for disease traits.The result of our study as follow:1. Collected 32 Fusarium oxysporum f. sp. Vasinfectums, The RAPD clustering showed that 32 species will be divided into 3 major categories, 5 sub-groups as in 7 physiological strain. By the way we screened high resistance cotton varieties 98134 and susceptible Xinhai 14(Gossypium.barbadence L. )2.Total 560 pair of SSR primers were used to select polymorphic markers from 98134 and Xinhai 14. There are 126 pairs of primers with polymorphism between the two parents, 119 pairs of primers were used to screen the F2 individuals, Using MAPMAKER/EXP3 .0 (the min LOD is 3.0 and the max genetic distanceis 50 c M.),a genetic map was constructed which contain 91 markers and 22 linkage groups covering a genetic distance of 1361.7 cM.3.The Fusarium wilt traits of cotton in F2:3 was showed to be normal distribution.The composite interval mapping(CIM) method was used to analyze the quantitativer tait loci(QTLs) of Fusarium wilt Resistance index taits based on F 2:3 family,4 QTLs of Fusarium wilt traits of cotton were identified,which located in linkage group 3 ,15,23a nd26, rspectively.The four of Fusarium wilt traits of cotton resistance related QTLs could explain,12.4%, 20.96% ,4.7%and 11.9 % of the phenotypic variation, respectively.
