Extraction,purification and bio-activity of polysaacharides,from the skin of Aloe vera Miller irrigated with sea water,were investigated.Crude polysaccharides,decolorred by carbon amorphous,were extracted by of water extraction and alcohol precipation.Response surface method was used to optimize the extraction of crude polysaccharides.Crude polysaccharides were advancingly isolated on DEAE-52 anion-exchange column,eluted with the deionized water,0.1 mol/L NaCl,0.3 mol/L NaCl,0.5 mol/L NaCl,1.0 mol/L NaCl,0.1 mol/L NaOH and 0.5 mol/L NaOH at a flow rate of 50 mL/h.Four fractions are obtained,named SAPS-A(deionized water),SAPS-B (0.1 mol/L NaCl),SAPS-C(0.5 mol/L NaCl)and SAPS-D(0.5 mol/L NaOH),which were 97.43%,1.45%,0.24%and 0.88%of the total amount of collected polysaccharides, respectively.SAPS-A was further purified on Sepharose-4B,one fraction was obtained, named SAPS-1 with 40±2kDa molecular weight.IR and NMR study of SAPS-1 demonstrated that the main skeleton of SAPS-1 wasβ-(1→4)-D-linked mannose with acetylation at C-6 of pyranosyl.The degree of acetylation of SAPS-1 is 40%.In a concentration-dependent manner,SAPS-1 was demonstrated to have antioxidant activities.Efficiency of scavenging activity of superoxide radical,inhibition of lipid peroxidation and scavenging activity of hydrogen oxide was 1/2,1/8 and 1/4 of Vc respectively.SAPS-1,a kind of mitogen,could enhance of nowhere which was spontaneously or which was induced by ConA.20μg/ml SAPS-1 could well promote proliferation rate of primarily cultured and ConA induced spleen cells to be 20.04%and 25.18%.Demnification of spleen cells was remarkable under UV,160μg/ml SAPS-1 was able to enhance protection and restoration of demnified spleen cells to be 26.24%and 43.56%.
