The Diversity Of Resistance To Jujube Witches' Broom Disease In Ziziphus Jujuba Mill. Cv. Junzao

Jujube witches' broom(JWB), a kind of vascular bundle system disease caused by phytoplasma, is one of the most destructive and infective diseases in Chinese jujube.In the former study, we have found that different lines of the same cultivar had distinct differences on JWB resistance. In this research, the JWB resistance of twenty-seven lines of Junzao - a high JWB resistance cultivar was studied. The aim is to screening the high resistance lines. Moreover, in order to further production application and gene engineering breeding, the characteristics of nutrition, morphology and genome were also studied.The main gained results were as follows:1 Screening 4 high JWB resistance lines from JunzaoFour high JWB resistance lines of Junzao were selected out using the method of grafting the healthy scions (Junzao) of the germplasm to be checked onto seriously diseased trees. On Zanhuangdazao rootstock, T7,T15,J2,J5,J6,J8 played high JWB resistance and on Fupingdazaorootstock, T8, T15, T16, J2, J10, J7, J8, J5 played the same resistance. There are four lines T15, J2, J5, J8 played high JWB resistance on different rootstocks.2 Different lines of Junzao played significant differences on JWB resistance According to apparent symptom, the different lines of Junzao cultivar can be dividedinto 4 kinds:â‘ Liable affected kind. The plant displayed symptom in the whole developing-periodâ‘¡Lingering affected kind. The plant apparent symptom only at the later stage of the developing period.â‘¢Lingering resistance kind. The plant only played resistance at the later stage of the developing period.â‘£Resistance kind. The plant never apparent symptomThe results determined by DAPI fluorescence method and PCR phytoplasm method are consistent with the apparent symptom investigation results.3 There exists no relativity in nutrimental indexes, morphologic indexes with resistance activity.By means of 16 lines (Junzao) nutrient indexes and morphologic indexes.analysis, we found that they have closed genetic relationship and there have not relativities between nutrient and morphologic indexes with resistance activity.There have not significant differences among nutrient indexes by rational-satisfactory degree analysis. The rational-satisfactory degrees of the resistance lines T15, J2, J5 and J8 are 0.60,0.48,0.67 and 0.58 and they are at a relative higher standard.4. There have significant differences on genome diversity among different Junzao lines.One hundred-four differential expression bands were screened out from different lines (Junzao) by using 10 pair primers. Among them, there have 2 special expression bands in resistance lines, 92 special expression bands in liable affected lines and 10 special expression bands in lingering affected lines and lingering resistance lines.
