Immunohistochemical method was used in detecting the localization of gabra1 in hypothalamus,pituitary and ovaries of Xiao Meishan pigs of newborn,30,60,70,90 days and Su Jiang pigs of newborn,60,120,150,180 days.The results showed that gabra1 positive particles were distributed in hypothalamus generally. A comparison of different age, the numbers of gabra1 positive particles in the period before puberty and after puberty were significantly less than puberty in hypothalamus . Gabra1 positive particles of pituitary were concentreted into pituitarium anterius cells. Gabra1 positive particles of ovaries appeared in granular cells; granular cells of all types of follicles. In ovaries gabra1 positive particles were expressed in all day, the difference was not significant difference.Quantified the gabra1 gene expression in hypothalamus,pituitary and ovaries of pigs at different ages.The RT-PCR results showed that: the levels of gabra1 mRNA was gradually increased from the newborn period to puberty and the levels was lower after puberty.The levels of gabra1 mRNA showed marked at difference days in Xiao Meishan pigs(P<0.05).In Sujiang pigs,the levels in hypothalamus was not significantly different between newborn period and 180 ages (P>0.05) and was significantly different among other days. The levels in pituitary and ovaries at difference days was significantly different (P<0.05).The levels of gabra1 mRNA in hypothalamus of Xiao Meishan pigs was significantly higher than that in Sujiang pigs at different ages(P< 0.05).Plasma reproductive hormonal changes were detected in pigs by radioimmunoassy.The results indicated that serum concentration of FSH,LH,E2 were high at birth, then declined and maintained at a low level. Concentration of FSH,LH,E2 rose quickly at puberty in Xiao Meishan pigs and Sujiang pigs. Concentration of P was low before puberty,and rose quickly at puberty.To sum up,our study were good for studying biological effectiveness of gabra1 at the onset of puberty and provide theoretical basis for illustrating animal reproductive mechanism and breeding of early-maturing varieties.
