Xiaomeishan pig is famous for its excellent reproductivity. Reproductive traits are important economic traits to pig, which directly influence economic benefit of swine production. Previous researches have shown that OPN(osteopontin) gene and BMP15(bone morphogenetic protein-15) gene were the candidate influencing genes for swine reproductive performance. With Xiaomeishan pig and Large White pig as the objects, this article contributed to genetic variations of both genes and their influence on swine reproductive performance. It also enriched the research of both genes and discussed how to apply the genes in the breeding process.1. According to the sequence of OPN gene published in GenBank, 6 PCR primers were designed to amplify the fragment. The result showed 1 microsatellite marker at P1. Tested the others with PCR-SSCP techniques, 3 mutations, 430T↓431G→A,473G→A, 402T→G, were detected at P3 and P5.2. With least square analysis, it was studied how different polymorphic loci of OPN gene influence swine farrowing performance, i.e. TNB(total number born) and NBA(number born alive). The result showed significant difference for both genotype sows at P1(P<0.05) with a trend of DF>AD>BD, significant difference for Xiaomeishan sows and for second and later parities Large White sows at P3(P<0.05), but no significant difference for both genotype sows at P5(P>0.05).3. According to the sequence of BMP15 gene published in GenBank, 2 PCR primers, P7 and P8, were designed to amplify the fragment. Their polymorphism was detected with PCR-RFLP techniques. The result showed no mutation detected at P7, 1 mutation at P8, i.e. T→A at 62 of exon2, which caused Bcu I-RFLP, and they were BMP15AA, BMP15AB and BMP15BB , respectively.4. With PCR-RFLP techniques, molecule scanning was done to BMP15 gene of Xiaomeishan pig and Large White pig. The result showed significant difference in their genotype frequencies and allele frequencies of BMP15, which was in accordance with reported findings.5. With least square analysis, it was studied how BMP15 gene influenced swine farrowing performance. The result showed significant difference between different genotypes for both Xiaomeishan pig and Large White pig in TNB(total number born) and NBA(number born alive), with a trend of AA>AB>BB. So conclusion was drawn that the allele A was good gene to improve litter size, and AA was a good genotype.
