Molecular marker-assisted selection (MAS) provides a very efficient tool for backcross breeding because it can offer rapid and precise selection of the targeted gene.The rice wide compatibility is a complicated trait. In traditional rice breeding of wide compatibility, it was identified by testing cross. With the developing of molecular and deeper study of wide compatibility, molecular marker-assisted selection (MAS) makes the selection to wide compatibility possible. The objectives of the study were to breed new sterile line and restorer line of wide compatibility gene (S5n)by selecting molecular marker J15 ,and identify the reliability of J15 by testing cross ,and the work what we done can supply a rapid and precise method to select new rice lines with S5n. The results are summarized as following:1,The wide compatibility gene (S5n) in rice variety 02428 were introduced into the rice maintainer line Zhen 5B,by the way of molecular marker-assisted selection (MAS),and then identify the wide compatibility of some individuals in BC1F1 population .The results are summarized: The wide compatibility gene (S5n) was enhanced the fertility of inter-subspecific hybrid, and compared to the average spikelet fertility of CK(Nantehao/Zhen 5B), genetic effects were in 17.10%~20.86%.2,The wide compatibility gene (S5n) in rice variety 02428 were introduced into SIR 2622, by the way of molecular marker-assisted selection (MAS), and then identify the wide compatibility and the restorer capability of some individuals in BC1F1 population .The result of wide compatibility identification was summarized :compared to the CK(Balilla /SIR 2622)the average spikelet fertility, genetic effects of the wide compatibility gene (S5n) were in 17.10%~20.86%. The results of restorer capability identification were as follows: the spikelet fertility of test-crossing offsprings in HL-cytoplasmic Male Sterility Indica Rice, Yuetai A and WA-cytoplasmic Male Sterility Indica Rice ,TianfengA was restored, and the spikelet fertility of test-crossing offsprings was about 80%,that is the normal level of self-cross.
