Brassica napus grains were taken as the materials.In this experiment we used paraffin section method to observe the oil distribution of the Brassica napus seeds with high and low oil content.Studing the changes of relative quality indicators and relative enzyme activity of Brassica napus seeds with high and low oil content during growth,we could discuss the formation mechanism of the oil content difference.Using different chemical regulation reagents in different times during growth,we could study the changes of oil content and protein content effected by chemical regulation reagents,and choice the best one.The results indicated:1.The method discussion of using paraffin section to observe Brassica napus seed oil distribution indicated that the best fixation was using K2Cr2O7 solution to fix materials in 15h. After embedding material in paraffin,we can dehydrate firstly and then use sudan black B to stain rape seeds slice.This method could show oil distribution in rape seed bestly.2.The observation of organization distribution about the Brassica napus seeds with high and low oil content indicated that:in the higher oil content seeds the staining region was broader,the dyeing was deeper and uniform and the edge was clearer than the lower ones.In the same rape seed the embryo cells were larger than cotyledon cells and the oil distribution was concentrated, but the oil occupied only half of the embryo cells space.The cotyledon cells is smaller,but the cells were intensive and contained more oil.3.The comparison studing of different(?)ty indica(?)of Brassica napus seed with high and low oil content indicated that th(?)R51's(?)est and the R75's oil content was ##原图像不清晰 lowest.Rape seed oil content and protein content were in a very signifi.cant negative correlation; oil content and soluble sugar had also had a negative correlation;1000-grain weight and soluble sugar content were in a positively correlated.4.The study about changes of formation mechanism along the Brassica napus seeds with high and low oil content fond:in the Brassica napus seeds with high and low oil content,the changes of the oil content,chlorophyll content,sucrose phosphate synthase of the peer,sucrose synthase of the seed and pyruvate kinase of the seed were consistent.The indicators' absolute value of the high oil content Brassica napus was higher than the low oil content Brassica napus.But the lysophosphatidic acid phosphatase reached the peak 50 days after flowering in the high oil content Brassica napus and reached the peak 45 days after flowering in the low oil content Brassica napus. The lysophosphatidic acid phosphatase has important influence on formation of the oil content difference.5.The study of the effect of oil content and protein content of the Brassica napus using different chemical regulation regents in different times indicated:spraying 5%citric acid after 33 days in the flowering could improve the oil content,but the protein content was declined at the same time.Overall,using chemical control reagents after 33 days in the flowering could increase oil content,but these treatment could slightly lower the protein content.
