Poplar is the most adaptive specie that distributes widely in the world.It is also the important specie that used in protection forest,soil-water conservation forest,environment greening and artificial forest.Large numbers of poplar clones by selecting can increase productivity greatly and take great roles in solving the national shortage of timber.This study used F1 generation group that hybridized by 84k for male and 101 for female as the experiment material,used BSA and RAPD marker,took preliminary selections of poplar fast-growing traits related genes linked by molecular marker.It provided basic molecular support for poplar high-yield breeding initially.Major results were as follows:1. The purity of DNA was in high level when the concentration of PVP in the extract was 0.5%,By this condition we can received integrity,high quality DNA,and can access stable PCR result.2. RAPD primer application had a high selectivity in the experiment,If annealing temperature and specific amplified products were higher in a certain temperature range,amplification was more stable.3. By screening polymorphism between fast DNA pool and low DNA pool which were established by F1 with 200 random 10-mer-primers, primer S47showed polymorphic between two DNA pools.One RAPD marker S47-1166 produced by primer S47(GGCACTGAGG)was observed in every plants of low DNA pool.4.The marker S47-1166 was cloned and sequenced.The sequence analysis revealed that S47-1166 has an 99% homology with the Populus tremuloides cytochrome oxidase subunit 1 (cox1) gene.
