Foot-and-mouth disease (FMD) is a highly contagious disease of cloven-hoofed animals, including cattle, pigs, sheep, goats, and many wild animals. FMD virus (FMDV) is a positive sense, single-stranded RNA virus of the family Picornaviridae, genus Aphthovirus. FMDV consists of a genome of approximately 8,500 bases surrounded by four structural proteins to form an icosahedral capsid. The RNA is translated as a single long open reading frame (ORF) into a polyprotein, followed by a series of posttranslational proteolytic cleavages to generate both the intermediate and mature structural (encoding VP4,VP2,VP3,VP1) and nonstructural viral proteins (2A,2B,2C, 3A, 3B,3C and 3D). There are seven immunologically distinct serotypes: O, A, C, SAT1, SAT2, SAT3 and Asia1, which contributes to difficulty in eradicating and controlling FMD. FMD is a worldwide disease and is endemic in regions of Africa, Asia, South America, and Europe. Chinese serotype Asia1 FMDV was first discovered in Baoshan County, Yunnan Province, in 1958. In the 30 years, Asia1/YNBS/58 was the only lineage of Asia1 FMDV since it was discovered in China. In April 2005, FMDV serotype Asia1 outbreak in Jiangsu Province, Gansu Province, Beijing and Hebei Province caused great damage to livestock industry. FMDV serotype Asia1 Asia1/JS/CHA/05 lineage, a different lineage from Asia1/YNBS/58, was descendent of Asia1/IND/1980, the complete genome information of FMDV would contribute to the development of new vaccines, diagnosis techniques and the understanding of FMDV pathogenesis.Here we report, the isolation and complete genome analysis of foot-and-mouth disease virus Asia1/1/YZ/CHA/06 isolate from pig, which belongs to lineage Asia/JS/CHA/05. In the same time, according to the complete genome of the FMDV Asia1/YS/CHA/05 isolate from bovine, which was previous sequenced in our laboratory, six cDNA fragments covering the FMDV Asia1/YS/CHA/05 complete genome were amplified and cloned into low copy number vector pOK12, 5'RACE was adopted to gain the authentic sequence of 5'end, and the full-length genomic cDNA of FMDV Asia1/YS/CHA/05 was constructed.
