In vitro maturation(IVM)oocytes are basic materials for modem embryo biotechnologies such as fertilization,nuclear transplantation and gene transfer. Although IVM of buffalo oocytes technique has been developed for a few years, the IVM oocytes quality are still not as good as the in vivo maturation oocytes. A major problem is that mechanism of oocyte maturation is not very clear.Here, the different expression genes in buffalo immature and matured oocytes were investigated,which could help understanding the mechanism of oocyte maturation in the molecular level.Using differential display reverse transcriptase polymerase chain reaction (DD RT-PCR)technique,we found nine differential express mRNAs between IVM oocytes and immature oocytes.The PCR productions of these nine genes were cloned to the pMD18-T vector and the recombined plasmids were sequenced.After compare with the Genbank or database using BLASTn tool,six of these genes were high homology with RPL31,LIMHOX,CAF1A,eIF4, RHOBTB2 and cep350 respectively.The differential displayed A4 fragment was not similar to any nucleartides in GenBank,so it may be a novel gene expressing in unmaturated buffalo oocytes.
