As at home and abroad on,plantlet in vitro and its roots are rarely described quantitatively in sterile conditions,plant hormon is also recognized as one of the key factors which affects the growth and photosynthesis of plantlet in vitro,the appropriate hormon assembly directly impacts on the commercial operation of plantlet in vitro.This study,which was in the use of image analysis technologies sterile dynamic monitoring of plantlet in vitro and its root growth,improving on-line testing on the basis of photosynthetic rate,has done some basic and pioneer discuss.The main content of this study includes the following:1.As the environment of plantlet in vitro growth is very special,it's difficult to growth and development of roots dynamic monitoring of the situation.Image analysis length of plantlet in vitro root is measured and described dynamically the growth and development under sterilized condition,is acquired through the analysis on the image from plantlet in vitro root.Analysis the reason for image analysis mass too small,gain corrective model:S=hr/L.The result from correlation analysis indicated that the plantlet in vitro image analysis length of root appeared a significant positive linear correlation with its manual length(R2=0.993,n=22,P<0.001).The logistic equation may fit for the observed value from the plantlet in vitro image analysis length of root. Image analysis length of Y and time X index equation is Y=1/(1/a+d(bX)),where a value means the biggest growth length of plantlet in vitro roots,and b value means exponential growth factor.2.As the environment of plantlet in vitro growth is different from the general of the daejeon plant,its growth and development situation is known very little.Image analysis mass of plantlet in vitro is used to measure and describe dynamically the growth and development under sterilized condition,was acquired through the analysis on the image from plantlet in vitro.The result from correlation analysis indicated that the plantlet in vitro image analysis mass,which obtained from two controls,manifested a significant positive linear correlation with its manual mass(R2=0.9108,n=29, P<0.001).The logistic equation may fit for the observed value from the plantlet in vitro image analysis mass.Image analysis mass of Y and time X index equation is Y=1/(1/ a+d(bX)),where a value means the biggest growth mass of plantlet in vitro,and b value means exponential growth factor.3.The growth of plantlet in vitro relies on photosynthesis to provide its own energy,photosynthesis of plantlet in vitro directly affects its own growth and development,and plant hormon is an important factor which affected photosynthesis of plantlet in vitro.The auxin assembly of 0.1mgl-1NAA and 2mgl-16-BA makes photosynthetic rate of Orychophragmus violaceus plantlet in vitro reach the maximum for 20.941 umol.kg-1.min-1,Carboxylation efficiency reach 0.029,and Dark respiration rate reach the maximum for 3.840 umol.kg-1.min-1;The auxin assembly of 0.1mgl-1 NAA and 2mgl-16-BA is the most advantageous for photosynthesis of Orychophragmus violaceus plantlet in vitro.4.The logarithm growth period of root in sterilized condition was acquired by analyzing the image from plantlet in vitro roots.The earliest occurrence time of roots is 10.65 days and growth rate during the logarithm growth period of roots reaches the maximum for 2.103cm/day,which resulted from 1/2 MS medium supplemented with 0.05 mgl-1NAA,0.5 mgl-1IBA and 0.003 mgl-1GA3.Through the plantlet in vitro root formation time and growth rate during the logarithm growth period of roots of comparison,The auxin assembly of 0.05 mgl-1NAA,0.5 mgl-1IBA and 0.003 mgl-1 GA3 is the most advantageous for root growth of Orychophragmus violaceus plantlet in vitro.5.The logarithm growth period of plantlet in vitro in sterilized condition was acquired by analyzing the image from plantlet in vitro.Orychophragmus violaceus plantlet in vitro grown very well and growth reached the maximum,the duration time of the logarithm growth period was 19.66 days,and the maximum exponential growth factor during the logarithm growth period was up to 0.913,which resulted from MS medium supplemented with 0.10 mgl-1NAA and 2.00 mgl-16-BA.Through in various auxin assembly of the duration time of the logarithm growth period,of exponential growth factor during the logarithm growth period,of the maximum growth mass of comparison,The auxin assembly of 0.10 mgl-1NAA and 2.00 mgl-16-BA is the most advantageous for growth of Orychophragmus violaceus plantlet in vitro.
