Environmental stresses,such as drought,salinity and flood have an significant impact on crop productivity.Wheat is the second important crop in China.With increasing population and decreasing arable land,Using the SNP of genes to search for the associativity bettwen environmental stresses and genes and to discover novel stresses-tolerant gene resources are not only important in theory but also valuable in practice for breeding stresses-resistant and high yield variety and promoting the development of agriculture.TaCCaMK gene is cloned from wheat,which activity depend on calcium and calmodulin.So far,this gene has been reported invoving in drought and salinity stresses-resistant.In this study,using three common wheat diploid relative-species,one tetraploid relative-species,Chinese Spring,a set of nulli-tetra lines(Chinese Spring background),five drought tolerance and three flooding tolerance as plant materials,we reported our work on the gene structure,chromosome localization,SNP of TaCCaMK gene EF hands domain,the correlations bettwen SNPs of EF hands domain and drought tolerance and flooding tolerance of TaCCaMK.The results were as follows:1) TaCCaMK contain 7 exsons and 6 introns.The number and the location of introns had no obvious changes in different genome varieties except for the size of the introns.The result revealed the conservation of TaCCaMK.2) The result of chromosome localization showed that TaCCaMK is located at chromosome 1A,1B,1D.The copy of TaCCaMK had been determined preliminarily through the SNP analysis of EF hands domain.which had at least six copies on wheat genome and two copies on chromosome 1A,1B,1D respectively,the two target sequences had only three base difference at the array lenth.3) During the evolution process of wheat,the sequence of EF hands doamin on each genome had specific variation.Of them,the variation on B genome was the most, contrarily,A,D genome was relatively conserved.The different degree of nucleotide variation of EF hands domain were detected on each genome,which was related to the defferent seletion power possibly. 4) An analysis on the affiliation of EF hands domain on three genomes based on the SNP in EF hands domain TX-copy-1 sequence showed that genome A and B had a near affinity.Genome D diverged from A and B largely.On the other hand,our result showed that the affiliation of EF hands domain of B genome from tetraploid relative-species and hexaploid wheat was closed,but which were distant from the B genome from diploid relative-species.5) The SNP of EF hands domain was abundant in two resistance accessions.A total of 14 SNPs and 5 InDels(insertion and deletion) were identified from 3 flooding tolerance and one Chinese Spring accessions,on average of one SNP every 402bp and one InDel every 1125bp;a total of 28 SNPs and 8 InDels were identified from 5 drought tolerance and one Chinese Spring accessions,on average of one SNP every 302bp and one InDel every 1005bp.This indicated that the SNP of EF hands domain was much more abundant in drought tolerance than flooding tolerance accessions.At the same time,we detected that the SNP of EF hands domain was much more abundant in B genome than A and D genome among the accessions.6) Analysis of haplotype revealed that the correlation bettwen EF hands domain from different genome and the properties of drought-resistant and flood-tolerant.Our result showed that TaCCaMK gene from A genome had certain correlation with drought tolerance,but had no obvious correlation with flood tolerance;the gene from B genome had obvious correlation not only with flood tolerance but also with drought tolerance;contrarily, the gene from D genome had no correlation not only with flood tolerance but also with drought tolerance.This indicated that the homologous gene of TaCCaMK gene from different genome plaied a different part against flood and drought,the homologous gene located at different genome involved in different stress response.
