Ligusticum chuanxiong .Hort is one kind of traditional Chinese medicine, it is mostly be used to cure blood vessel disease of heart and brain, pulmonary disease, nephropathy, liverish, neuropathic ache, and so on. It has a centuries-old history with 1500 years. DU Jiang-yan city in Sichuan Province is the trueborn producing area, whereas, the seed of Ligusticum chuanxiong .Hort were lack of uniform and canonical management, the seed were thread and thrum in many different breeding bases, so all the upwards factors influence its yield and quality seriously. Otherwise, the root rot and miller of stem node are two most important plant diseases and insect pests, they are also serious influences. DU Jiang-yan was chosen as the test place, compared and analyzed the differences of main agricultural characters, dry-weight, yield and principal component of Ligusticum chuanxiong Hort from five breeding areas, in order to get the best seed, and provide the academic proof to the breeding and planting through correlation analysis between single weight and main plant traits of Ligusticum chuanxiong Hort. besides, built the fast breeding system, heighten breeding coefficient, shorten breeding time consequently, and through the research of a series of affect in the progress of tissue culture, establish the foundation of secondary metabolite production, the results as below:1,there was a big difference among five LING ZHONG ,on the side of agronomy characters, the height have inapparent difference in the fade period and harvest, but the LING ZHONG from Tai'an(one town of DU Jiang-yan city) was better than the others, the height in December and May was 48.79cm and 44.48cm respectively, but the number of leaves had significant difference, LING ZHONG from Tai'an have the most leaves, the number of leaves of Ba Jiao(one town of Shi Fang city) LING ZHONG was the least, in September ,the seed began tiller, the number of tiller reached maximum in the fade period, it also had obvious difference, the number of tiller of volatile exceeded 82.1% of Ba Jiao LING ZHONG in the harvest.On the side of dry-weight accumulation, the LING ZHONG from Tai'an had the first-rank proportion between rhizome and leaves in the fade period, the ratio was 1.94,in the harvest, the proportion among leaves ,stem and rhizome of Tai'an LING ZHONG was maximal (1:1:4),the average dry-weight was 106.17g/plant,and the proportion among leaves, stem and rhizome of Ba Jiao LING ZHONG was 1.58:1:5.54,but the average dry-weight was just 69.55g/plant,so the proportion could not be high, and the harmony development of different organs should be thought much .On the side of yield , the yield of Tai'an LING ZHONG was 5057.4 kg/hm2 which was more than others and the difference was significant, compared with Ba Jiao LING ZHONG, the production increased 33.2%.On the side of content of principal component, the content of total ferulic acid and volatile oil of Tai'an LING ZHONG were more than other seed, the content were 0.227g/100g and 1.527g/100g respectively. the seed which came from Shui Mo(one town of Wen Chuan county) had the least content of total ferulic acid and volatile oil ,the content were 0.181g/100g and 1.453g/100g.otherwise,the LINGZHONG from Ba Jiao had the most content of total alkaloid(0.534g/100g),the content of total alkaloid of Tai'an LING ZHONG was next (0.468g/100g),because of the high yield and the content of total ferulic acid ,volatile oil and total alkaloid, the three content of principal component of Tai'an LING ZHONG on the unit ground area were the most (12.05kg/hm2,77.27kg/hm2 and 23.7kg/hm2), the result showed that the LING ZHONG from DU Jiang-yan was better.2,the yield was related to height, R/T, the number of tiller and leaves, stem wide, leaves weight and stem weight ,the fitting measures should be adopted to increase R/T, the number of tiller, leaf weight and stem weight. and reduce height, stem weight and the number of leaves accordingly in order to heighten the yield. the number of tiller, stem wide and weight of leaves in harvest should be the main factors in progress of seed breeding and high yield planting.their stepwise regression mode was Y=86.94-3.28X1+143.60X6-4.69X7 +2.93X8-0.89X9+424.96X10+2.64X11-1.68X12.3,the best season for getting stuff in the process of tissue culture of Ligusticum chuanxiong .Hort was April, stem node was the best explant and start-up time of callus tissue was 5 days, inducement rate reached 88.3%, 0.1%HgCl2 was the best disinfector for stem node ,it's effect was better than 2%NaC103 greatly, the best treatment time was 6 seconds, both the rate of pollution and death was 0,and there was no any damage to explant.4,the effect of NAA was better than 2,4-D on the side of callus tissue, although 2,4-D had the strong ability to advance the formation of callus tissue, the character of callustissue was bad ,it was white and soggy, 2,4-D restrained bud formation mightily, with concentration increased, the callus tissue was dead easily in the anaphase.2.0mg/L NAA was better, the callus tissue was loose and distinct green in the anaphase, it was good for differentiating next step.5,The main factor in the step of stem node callus tissue inducement was sucrose, but in the step of differentiation, 6-BA was the main factor, the best inducement culture medium was MS+1.5 mg/LNAA+0.5mg/L6-BA+40g/Lsucrose,inducement rate was 85%,and the best differentiation culture medium was MS+2.0mg/L6-BA+0.2mg/LNAA +20g/Lsucrose, differentiation rate was 86.67%.6,sucrose was better than glucose in the carbon resources, the callus tissue was eugonic and loose in the culture medium with sucrose, 30g/L sucrose was the best, the wet-weight and dry-weight ratio were 365.21% and 265.21% respectively. 10g/L glucose was better in the different concentration of glucose, the wet-weight and dry-weight ratio were 240.15% and 140.15%.7,the best organic additions was 450mg/LCH,the callus tissue inducement rate was 58.33%,and the differentiation rate was 61.67%.8,the best rooting medium was 1/2MS+0.5mg/LNAA+30g/Lsucrose,the rooting rate was 100%, the tufty seedling had more acerose root with white color ,the length of root was about 5.5cm,the average number of rooting was 7.2.
