In an agriculture setting,the use of transgenic technology to increase crop production is an economic and simple way. At present, the area and types of transgenic crops is increasing year by yaer in domestic and foreign countries.And it obtains the good economic efficiency.The fact proof that the transgenic crops' using prospect is very widespread.At the same time,along with transgenic crops'area unceasing expansion,the humanity also give more attention to the safety of transgenic crops. Reaching for many years,we have transformed the wheat xi'nong-1376 with the leaf senescence-inhibition gene PSAG12-IPT.For it can be used in the future breeding work, with the animal model of white rats we analysed the edible security of the wheat xi'nong-1376 with the leaf senescence-inhibition gene PSAG12-IPT; Through the PCR examination to the rhizosphere soil, rhizosphere animalcule, rhizosphere weed, pollen excursion and the straw residuum of the transgenic wheat we analysed it's entironment security;For selecting the plants which may receive the PSAG12-IPT gene fastly and effectively,we analysed the suitable hygromycin concentration for selecting the transgenic wheat and barley.the test result is that:1.The small white rats feeding transgenic wheat xi'nong-1376 with PSAG12-IPT gene and common wheat xi'nong-1376 were studied separately. The weight change,baby quantity haematology and biochemistry indexes, organic histological maps of liver, kidney, intestines, lung, cerebra and testicle of parents and baby generation rats of the two groups were analyzed. It indicted that transgenic wheat xi'nong-1376 with PSAG12-IPT gene did not have obviously bad influences to small white rats health and might be safety to human health.2.With the hygromycin concentration 0,50,80,110, 140mg/L and 0,20,40,60,80, 100mg/L we tested the wheat xi nong-979,ning chun-16,zheng-9023 and barley B1202,U202,yun yin-Num1 separately. It indicted that 140mg/L can restrain non-transgenic wheat to grow and 80 mg/L can restrain non-transgenic barley to grow. We selecting the ransgenic wheat zheng-9023 generation1 and barley B1202 generation1 with the hygromycin concentration 140mg/L and 80mg/L, PCR result provide that the selection efficiency are 87.6% and 77.3%. The result indicated that Using hygromycin to select transgenic wheat and barley is feasible. 3.PCR analysed the rhizosphere soil, rhizosphere animalcule, rhizosphere weed and the straw residuum of the transgenic wheat.Also PCR analysed the wheat xi'nong-1376,zheng'yin-9023;barley yun'yin-I,yun'yin-III and the oat which selected by the hygromycin.The result indicted that PSAG12-IPT gene have not spreaded to the environment.
