The Correlative Study Of Wheat Ramosa Gene And Branched Spike

Apical meristem determines inflorescence architecture in Gramineae. Inflorescence architecture shows that meristem number,location,action, so the appearance of Gramineae plants is affected by inflorescence. Branched spike is abnormal type in Wheat ear. The main character is there are a lot of spikelets in principal axis. It can increase the number of spikelets and grains per ear. The study on phenomenon of branched spike not only favors to show the theory of Morphological Development in wheat but also has potential application value.Therefore, the study of the genetic mechanism in wheat inflorescence can rich the theory of inflorescence's Morphological Development and applied in high-yield production.Based on the study of our lab on chromosome mapping of branched spike gene , 7 SSR makers were selected on 2A and 2D。Two of them have polymorphism in the two parents by electrophoresis. Wmc453 and wmc819 were applied in the analysis of 96 individuals with branched-spike in F3 population. The result was that branched spike gene was linked to wmc453 on 2A chromosome, the genetic distance was 14.06cM . so it increased the linked maker of branched spike gene on 2A chromosomeMeanwhile ramose3 was cloned according to homologic sequences. Ramosa3 was analyzed by biology information. The result showed that RA 3 encoded 219 Amino acid sequence .The amino acid sequence deduced from the nucleic acid sequence had the respective similarity of 87% with maize,85% with rice,82% with Pennisetum glaucu and 86% with Setaria viridis. Domain analysis of the predicted amino acid sequence showed that Wheat RA3 encoded Trehalose-6-phosphate phosphatase. RA 3 is TPP family and can hydrolyze Trehalose-6-phosphate phosphatase.Whether Branched Spike gene in wheat confirmed by Genetic mapping is the same gene as ramosa2 or not? We found that nucleic acid sequence and Amino acid sequence were similar by Comparing between Branched YUANZHUI Wheat, ordinary wheat ,FEN33 and CS . The result informed that ramosa2 could not arose branched Spike. We produced ramose2 probe for southern blot and hybridized in 2A,2B,2D,QINBEN,CS,FEN33.The result is that ramose 2 is the same between 2A,2D,CS,and FEN33.So branched Spike gene is not ramosa2.
