Paraffin section method and light microscopy techniques were used to study on anatomical structure of seedling's primary vascular system of Glycine soja Sieb. et Zucc.for discussing the problem of seedlings joint of primary vascular tissue of G. soja Sieb. et Zucc. , and the correctness of Cotyledon node zone (CNZ)-theory was to be proved by comparing G. soja Sieb. et Zucc. with Glycine max (L.) Merr..The results showed that the CNZ of G. soja Sieb. et Zucc. is longer. There is mesarch tetrarch siphonostele in the lower part of CNZ, and meristematic tissue in the upper part of CNZ. From beginning to end, there is pith in the whole part of CNZ .There are two general joint types of primary vascular tissue of dicotyls seedling. One type has two transition regiones, and the other has a CNZ-root transition region and a CNZ-stem connection region. The joint type of primary vascular tissue of G. soja Sieb. et Zucc. seedling belongs to the latter. In addition, two–trace and unilacunar cotyledonary node in G. soja Sieb. et Zucc. seedling was observed.It was suggested that the CNZ type of G. soja Sieb. et Zucc. belongs to Basipetal shifting type.
