Distribution Of MDRV In Muscovy Ducks Detected By Immunohistochemistry Methods

84 10-day old healthy Muscovy ducks were infected with Muscovy Duck Reovirus(MDRV). The distribution of virus and its tissue tropism were detected by immunohistochemistry at 6h,12h,24h,48h,60h,72h,84h,108h,144h,204h,13d and 16d after infection. The purpose of this study is to explore the pathologenesis and early diagnosis. The research could also provide theoretical evidence for the collection of pathologic materials.The results of detection showed that:6 hours and 12 hours post infection,all organs were negative;24 hours post infection, liver, cecum,ileum and cerebrum showed positive cells first; 144 hours post infection,all organs were almost strongly positive;204 hours post infection,spleen,lung and thymus kept the strongly positive reaction while the positive reaction of other organs began to decrease; Kidney and Fabricius' bursa were weakly positive during the infection. The distribution of positive cells in different organs was of the characteristic from stroma to parenchyma and from blood vessel to cells.The target cells of MDRV were as follows, Liver cells, Endothelial of blood sinus,Kupffer's cells in Liver; Lymphocyte,macrophage in Spleen; Epithelial cells of renal tubules,Podocyte,Capillary endothelial cells in Kidney; Cardiac muscle cells, Capillary endothelial cells in heart; Epithelial cells of alveolus, Dust cells and Capillary endothelial cells in Lung; Thymic lymphocyte and macrophage in Thymus; Lymphocyte in Fabricius'bursa; Goblet cells, Lymphocyte and Capillary endothelial cells in Cecum; Lymphocyte, Epithelial cells of enteraden and Capillary endothelial cells in Ileum; Oligodendrocyte and Capillary endothelial cells in Cerebrum and Cerebellum. Positive lymphocytes could also be seen in Liver,Kidney,Heart,Lung and other tissues.The most strongly tissue tropism of MDRV was Liver, Cecum and Spleen,and these tissues were the target organs of MDRV.
