Productivity is one of the most important indicators in pig production industry. Enhancing pig reproductive ability will generate huge economic effect to pig breed industry. The GnRH gene, GnRH receptor gene and EPOR gene are important genes that are closely related to reproduction trait and they are principle candidate genes for reproductive ability.In this experiment these three genes are cloned. The sscp method was carried on with the important regions of these genes of varied pig breeds. Gene expression patterns of EPOR are constructed with three female large white; Association gene expression patterns of GnRH,GnRHR,FST,FSH,FSHR,LH,inhibin(a),inhibinβ(a) ,inhibinβ(b) which are of principal genes in the hypothalamus-pituary-gonad axis are also constructed with these three pigs. The following result are obtained:1. The majority sequences of GnRH gene and GnRHR gene are cloned2. No mutation is found on the coding region of these three genes.3. Two possible potential functional polymorphism sites are found at the 5'flanking region of GnRH receptor gene. One is an insert mutation within which a GAGA BOX exist. Another is a point mutation causing a transcription regulating protein IRF-1 binding sequence. These two sequences are already proved playing crucial function with transcription efficiency in many genes. The analysis indicates that for the insert mutation, there are three genotypes: AA, AB, and BB. The genotype Frequency exhibit great discrepancy varying according to the boar breed. The min-pig has very significant discrepancy compared with other breeds. The IRF-1 mutation exists only in min-pig group and there are only two genotypes discovered: AA and AB.4. According to the expression patterns of EPOR gene it can be speculated that this gene is widely distributed in pig tissues.5. According to the expression patterns of the hypothalamus-pituary-gonad axis major genes it can be concluded that there are no outside hypothalamus-pituary-gonad axis interaction system of these genes. These genes might have other effect within the pig tissues except for regulating reproduction cycle.
