In this paper, stems of elite Torreya grandis'merrillii'and seeds of T. grandis plant were used for ex-vivo, cultivation of seedling rootstock as well as the technology of shoots-graftingin greenhouse. The inducing system for axillary bud of T. grandis'merrillii'and the technology of shoots-grafting in greenhouse were established preliminarily. The main results obtained in thisstudy as follow:1. There are great influences on the effect of induction for axillary bud. As for collect time, collect locum and treatment are different. The axillary bud that was collected in April was the most easily induced from no-defoliation stem and the inductivity of axillary bud induced from crown of T. grandis was higher than that of T. grandis'merrillii'.2. It is concluded that The synergistic action of multi-hormone was the best method to the induction of axillary bud from the study for ex-vivo.The inductivity of axillary bud were 57.57%, 50.85% and 70.45%,when the stem was cultivated on B5+BA0.5mg·L-1+NAA1.0mg·L-1, B5+KT0.5mg·L-1+NAA0.8mg·L-1 and B5+BA0.1mg·L-1+NAA0.5mg·L-1+IBA1.0mg·L-1. The axillary bud that was induced could be cultivated to strengthen on MS+BA1.0mg·L-1 +NAA0.1mg·L-1. The seeding that was healthy and non-root grew 4cm after 20d. The best culture medium that was used for induction of callus was B5+2,4-D1.0mg·L-1+BA0.1mg·L-1 and the inductivity was as much as 84.67%.3. The accelerating germination rate was as much as 85% when the seed of T. grandis was planted in the double-layer plastic-cover greenhouse.It is more than mono-layer that is 58% and direct seeding membrana tectoria that is 47%. There are obviously different in 0℃, 5℃, 10℃, 25℃when the seed of T. grandis was germinated. The germination rate was 0% when it was treated at 0℃after one month and 96% when it was treated at 25℃.4. The most suitable rootstock for shoots-grafting was the immature rootstock that was three months old. The survival rate of grafting was up to 86.67%. It was 68.33%, 31.67% and 6.67% when the immature rootstock was planted six months, one year and two years old.5. It is favorable for grafting shoot to survive that tissue culture rootless shoot was hardened with bottles open in advance. The highest survival rate was 83.33% when the hardiness time was 6d old. However, there are only 11.67% when the grafting shoot was not hardened. 6. The shoots-grafting could be done in average year. The survival rate can reach to 60% above from February to December. The side grafting method was the most beneficial for the survival of grafting plant and the survival rate of the rootstock that was non-beheading was 86% comparing with 10.33% that was beheading.
