Rice, as one of the major food, the safety of transgenic rice has been a problem of concern to the people. The milled rice endosperm is part consumption of rice, if positioning in the endosperm of exogenous gene deleted, it can be completely resolved the issue of food safety of transgenic rice. The purpose of this study is use Cre / loxp sentinel delete system to delete exogenous gene in the endosperm of transgenic rice positioning, build a highly efficient transgenic rice endosperm exogenous gene deletion system. To achieve this purpose, rice endosperm-specific expression promoter Gt1 use for regulation the expression of Cre recombinant enzyme and the gus expression as a reporter gene, designed two lines to verify the feasibility of this study. For one line ,foreign genes in transgenic rice root, stem and leaves of not deleted,and deleted in the rice endosperm, this study constructed a corresponding vector pCALHGt1CGUS , transformation through Agrobacterium rhizogenes and obtain 202 cloning transgenic rice plants. 146 cloning survival. Molecular test showed that the majority of exogenous gene has been integrated into the rice genome, gus gene expression in the organization of chemical detection roots, stems, leaves, embryos, endosperm are not infected with color, RT-PCR detection In progress. Another line , in the root, stem, leaf, and other organizations the gus gene expression when the exogenous gene was not deleted ,in the endosperm of exogenous gene deletion and gus gene did not express, the corresponding vector pCLHGt1CreAGUS is being constructed.
