Moniezia benedeni is composed of scolex, neck and strobila, it has a series of changes in the shape. The changes of Moniezia benedeni in the shape, be that the gene mounts selectivity in time or space express, the results of gene differential displayed. Using the molecular biology experiment technology, selecting the correlated genes, takes the pattern biology by Moniezia benedeni, is helpful to the understanding higher biological organ growth the molecular mechanism.1. To make clear structure stained specimen of Moniezia adult tapeworm, observe its shape structure, summarize its shape change rule, the study sampled the Xinjiang Moniezia benedeni as experimental object, use the hematoxylin dye solution, carries on the dyeing to the Moniezia adult tapeworm, observe the stained specimen under the microscope. The result indicated that Moniezia benedeni from neck to gravid segment there are two sides vertical excretion tube, and the tube is thicker gradually. The reproductive system is appeared in immature segment, the female and male reproductive organ growth mutilation in mature segment. The male reproductive organ degenerate in the first in gravid segment. Later the female reproductive organ degenerate, then the gravid segment fill with the wombs.2. To select the differential displayed genes between the immature segment and mature segment of Moniezia benedeni, the study sampled the Xinjiang Moniezia benedeni as experimental object,according to the stained specimen of Moniezia benedeni, select the immature segment and the mature segment. The mRNA differential display reverse transcription PCR method (DD-PCR) was used to analyse and screen the differential displayed genes.Three anchor primers and eight arbitrary primers were selected. The follows could be done, RT-PCR,analyzing with electrophoresis, identificating and isolating the differential display bands, reamplificating,cloning with pMD18-T, Northern blot analyzing, sequencing and searching by Blast software. The results have eight differential displayed ESTs. SeqA,SeqB,SeqC,SeqG were differential expressed ESTs in mature segment of Moniezia benedeni. SeqD,SeqE,SeqF,SeqH were differential expressed ESTs in immature segment of Moniezia benedeni. The identities between the partial sequence of SeqE with Taenia asiatica mitochondrion and Taenia crassiceps mitochondrion genes are relatively high. The whole sequences, the localizations and the particular biological functions of them will be studied subsequently.
