Shellfish culture of the short food chain, the settling strongly, a good foundational seedling and breeding, and low cost has become one of the important pillar industry of mariculture in China's coastal areas. The triploid shellfish has several advantagements including poor fertility, rapid growth and better quality ,which becomes the emphasis of shellfish culture. Triploid culture has been applied main economic shellfish such as Pacific Oyster industrial production. 6-DMAP has been widely used in triploid culture due to its low toxicity, high effect and low cost among various methods. It can not ignorance that 6-DMAP could remain in environment, if not well treated, come into ocean will be harmful to marine biology, and it can be bioaccumulate and biomagnify through food chain (web), eventually threaten human health, and few reports concern about the safety of 6-DMAP. In this article, the safety of 6-DMAP is assessed by in vivo and in vitro assay. And it is found that:(1) fertilized egg and early embryonic development of Pacific oyster is effected by 6-DMAP and it adverse to development and growth of oyster. Exposed to 600 mM 6-DMAP for 15min, the ultrastructure of fertilized egg and early embryo is effected, mitochondrion slightly deform, parts of mitochondria cristae is dissolved and lower electron density zone is found; more yolk appear and electron density is higher, some yolk deform, there is a few lacuna yolk deform; endoplasmic reticulum slightly swelling, some ribosome desquamate, part of nuclear membrane deform slightly dissolve; 2-8 cells stage nuclear membrane slightly, endoplasmic reticulum swelling and vacuolization, mitochondria cristae apparently dissolve.(2) 6-DMAP could loose tight junction of cells, increase cell permeability. Exposed to 6μM, 300μM, 3000μM 6-DMAP, the value of Caco-2 cell significantly reduce with exposed time. (3) 6-DMAP may have estrogen like activity. MCF-7 proliferation are significantly enhanced by 93.75μM, 187.5μM, 375μM, 750μM, 1500μM 6-DMAP, and 3000μM 6-DMAP greatly enhance MCF-7 proliferation.(4) 6-DMAP may have potential carcinogenicity. DNA synthesis of Balb/c3t3 A31 are significantly inhibited by 500μM 6-DMAP, 1000μM 6-DMAP greatly inhibit DNA synthesis of Balb/c3t3 A31. DNA synthesis of Balb/c3t3 A31 reduce with concentration of 6-DMAP.(5) Nerve differentiation is promoted by 6-DMAP, meanwhile inhabit the activity of AChE, effecting nerve conduction and regulation. Exposed to 1250μM 6-DMAP neurite growth of PC12 is no difference with NGF's. but treated by 500μM,750μM,1000μM,1250μM 6-DMAP, the AChE activity of PC12 is inhibited, and the inhibition coefficient increase with concentration of 6-DMAP.In a conclusion, it is suggested that the cytoarchitecture and cell division is effected by 6-DMAP, having estrogen like activity, effecting nerve conduction and regulation. The mechanism of 6-DMAP should be further studied. And it is suggested that 6-DMAP should be recovered and reused in order to protect environment and human healthy in the future.
