Agrostis stolonifera L.is one of the most widely applied cool season turf grasses in China.To determine genetic diversity and relationship of Agrostis stolonifera L.resource, eighteen wild populations and a cultivated population(comparison) in Guizhou was screened by morphological diversity,peroxidase isoenzyme variance and inter-simple sequence repeat(ISSR) markers in the present study.Meanwhile,in comparison of Putter, we choose fine wild accessions to study their physiological change under heat and drought stress in summer.The results showed:1.All the morphological indexes showed abundant variance,the CV was from 17.16% to 34.3%.The Hs was 1.9991,and the Hin was 1.848,the HBetween was 0.1511,it indicates that the genetic variation among populations(92.44%) was greater than that within populations(7.56%).The diameter of the erect shoot,the leaf width of the erect shoot,the leaf width of the creeping shoot,the length of the median internode and the leaf length of the erect shoot were the main composing of morphological diversity.The result of cluster analysis was disaccorded with Agrostis stolonifera L.'s geography distribution in Guizhou.2.we detected two peroxidase isoenzyme points:Pod-1,Pod-2.The Pod-1 included 8 bands,RF were:0.031,0.046,0.065,0.088,0.111,0.131,0.163,0.183,it indicated there were four alleles:Pod-1a,Pod-1b,Pod-1c,Pod-1d,and Pod-1a,Pod-1b,Pod-1c were the main alleles:The Pod-2 included 8 bands,RF were:0.644,0.667,0.699,0.752, it also included four alleles:Pod-2a,Pod-2b,Pod-2c,Pod-2d,andPod-2c was the main allele.Based on the DICE genetic similarity,accessions could be divided into 4 groups.3.9 out of 60 pair primers produced total 81 bands,and 66 of them were polymorphic. Statistic analysis showed that the percentage of polymorphic loci(PPB) was up to 50.21%. The populations could be clustered into three groups based on GD.Correlation analysis found a significantly positive correlation between geographical distance and genetic distance(r=0.768,0.05>P>0.01),and this is persuasive evidence to support the conclusion that there was a significant correlation between genetic component and the geographic distribution of Agrostis stolonifera L resource. 4.Under heat and drought stress,every accession's physiological change showed follow rules:chlorophyll a,chlorophyll b and total of the chlorophyll contents had a descending trend;the relative permeability of cell membrane augmented;the contents of free proline accumulated gradually.This experiment applied membership function method, apprised their different tolerance to stress,they showed a difference as this:LPS3>BSE4>BNW2>BNW1>Putter>LPS5.
