Glutathione - S-transferase Pi(GSTP1) is an interiorly important detoxicated enzyme of mammals,involving in the maintaining balance of redox state.Research showed that glutathione-S-transferase in cells was involved in oxidative stress injury in the process when cells were stimulated by the heat,removing the heat stress free radicals.We choose GSTP1 gene as candidate gene which affect or contral heat stress of draiy cow.The association between bovine heat stress injury and SNPs which used sequencing of GSTP1 gene was studied.The results were as follows:The complete sequence of GSTP1 gene was sequenced,is 2869 bp.35 SNPs of GSTP1 gene was detected in 2 cattle breeds incluing 30 Hostein draiy cow and 15 LuXi yellow cattle.19 of 35 SNPs were reported by us.part of the SNPs located in intron,SNPs located in CDS belong to silent mutation except for A2696G,resulting in changes of amino acid(V181M) in LuXi yellow cattle.3 SNPs including G17C,T326C and A2696G was studied by PCR-RFLP in 120 hostein draiy cow.It is showed that 2 allele(denoted as G and C) and 3 genotype(denoted as GG,GC and CC )was detected in G17C loci,the frequency of 2 allele and 3 genotype was 0.4791,0.5209,0.2583 ,0.4417,0.3000 respectively.2 allele(denoted as M and N ) and 3 genotype(denoted as MM,MN and NN)was detected in T326C loci,the frequency of 2 allele and 3 genotype was 0.8917,0.1083, 0.8167,0.1500 0.0333,respectively.There is not digestion polymorphism in A2696G loci,but 2 allele were found in LuXi yellow cattle population,and the frequency of 2 allele was 0.1286 and 0.8714.GLM analysis for the effect of genotypes on index of heat torlerance(IHT) demonstrated that differect genotypes was not significantly associated with rectal temperature and potassium concentration of red blood cells(P>0.05).Tissue expression difference of GSTP1 gene under normal and seriour heat stress condition was detected by RT-PCR.Analysis showed that GSTP1 gene was expressed in heart,lung,kidney and spleen under normal and heat stress condition,and there was not significant difference in different tissue(P>0.05).However,there was significant difference in liver under normal and seriour heat stress condition.(P<0.05).
