we studied high oil content in the Brassica napus L 7218AB,YU-118 as experimental materials to develop a double-haploid plants for quality analysis with microspore culture technology.We can choose a high oil content and stability of the Brassica napus.The main results are as follows:1 Microspore culture of 'yu -118',7218AB,4666,4619 as experimental materials,And the results showed that the embryo production of'7218AB','4666' were 60%,37.5%respectively,but that of '1489' '4619' were only 10%,13.3% respectively;Significant differences in embryoid yield among plants of the same donor material were observed,4666-10 the highest embryo yield was obtained with 10.5 embryo per bud;4666-3 the relative lowest embryo yield was obtained with 0.05 embryo per bud, the ratio of embryos difference 10.45 embryo per bud.These results suggested that the genotype was an important factor of embryogenesis in the process of microspore culture in Brassica napus.2 the Brassica napus L 7218AB,YU-118 as experimental materials,every two days to select the length of 2.5~4 cm in bud in The same material the same plant,using the same conditions for the cultivation of microspore culture,In the first two materials sampling(That is,two of the beginning of flowering )for Microspore culture.The rate of embryos of '4666-1' is 10 cfu/dis,4619-1 with a lowest yield;To third when the sampling(about one week in initial bloom stage ),the ratio of embryos of '4666-1'and '4619-1' is 32.25 cfu/dis which The embryo has reached the highest rate;To the fifth sampling(fhll-bloom stage),The rate of embryos are decreased more.The results show that the effects different sampling time exerted significant inductive effect on embryoid formation;These results suggested that the Microspore culture of optimum period is 4~12 day after early flowering stage.3 The double haploid(DH) population derived from restore lines 'Yu118', '7218AB' by microspore culture were constructed successfully including 70 and 50 lines respectively in 2006;The double haploid(DH) population derived from restore lines '4666','4619 'were constructed successfully including 300 and 100 lines respectively in 2007.4 the normal diploid of'7218AB','Yu118'which use DH0 lines through receptacle culture were studied in this paper in 2007,Adventitious bud inducing is 56;Under microspore culture is not performing well and comparison of valuable material,the receptacle directly budding as remedies and methods.These results suggested that the best incretion proportion for direct bud organogenesis was 5.0 mg/L 6-BA and 0.5mg/L NAA.5 The double haploid(DH) population derived from restore lines 'Yu118'and'7218AB's Oil content of variation and distribution of statistics were Statistics, These results suggested that the variable amplitude of 7218AB with high oil content is 38.95~48.19,Coefficient of Variation is 5.28,more than 47 percent is 2,achieve the highest 48.19%in the others;The double haploid(DH) population derived from restore lines 'Yu118' with high oil content is 36.86~49.21,Coefficient of Variation is 6.9,more than 47 percent is 9,achieve the highest 49.21%.The other major quality traits have shown a certain degree of genetic variation,the largest variation appeared in erucic acid content,glucosinolates content and stearic acid content,and followed by oil content, oleic acid content and protein content.The variation of inolenic acid content and palmitic acid content was the least.6 18 DH lines whose oil content were higher than 44%in 2007 were checked again in 2008,18 lines in 16 lines of the oil content of improved,increase in oil content of 0.07~3.35 percentage points.Access to oil content of 48 per cent of the plant has four,the highest oil content reached 48.67 percent.High oil content of DH lines as the Brassica napus portfolio to provide material support.
