Cotton is not only primary fiber crop, but also important oil and protein plants inferior to soybean in China. Rise and fall of cotton production is crucial significance for country economy. However, there are many problems for the cotton traditional breeding technique. The main reason is the higher requirement of comprehensive character, including yield, insect-resistant, disease-resistant, stress-resistant, fiber quality of cotton cultivars, along with economical progress. Combination of cotton biotechnology and traditional breeding technique is a necessary approach for cotton breeding. At present, transgenic technique applied to cotton genetic transformation includes particle bombardment, Agrobacterium-mediated transformation and pollen tube passageway among many transgenic techniques, the former two technique need to establish genetic transformation system. Therefore establishment of steady and high efficient plant genetic transformation system is precondition assuring abundant filtered transgenic colony.Sea-island cotton cultivars that have some excellent characteristic, such as high fiber quality, outstanding disease-resistant, greensickness-resistant, is important resource used to cotton quality improvement, fastness inheritance and heterosis. In this, island cotton somatic embryogenesis and plant regeneration were primary involved. And the result is as follows:1 Twenty eight elite genotypes were selected for comparison of their regeneability using IBA+KT, NAA+KT and 2,4-D+KT hormone combination for the fist time. The results showed that three varities, namely, 'junhai 1','xinhai 3'and'GZZ488'have predominant ability of somatic embryogenesis, which got loose and flaxen callus within one month.2 Efficiency of callus inducement using different hormone and different hormone combination were studied. Efficiency of callus inducement using 2,4-D+KT hormone combination was established. Texture and state of callus were close to state of differentiation using different method, including changing variety and concentration of hormone, doubling KNO3 altering sugars, accretion, low temperature. However, efficient result was not obtained by reculturing. 3 Three Xinjiang sea-island cotton, 'Xinhai 17', 'Xinhai 14', '85H', was studied on shoot organogenesis and genetic transformation. The results were as follows: in vitro plant regeneration of meristem shoot tip is better than other organizes. The rate of 90.5%.were obtained. while applying 6-BA at 0.5 mg/L, frequency of inducing shoots reach maximal; whereas number of budding shoots in per explants reach maximal with 6-BA at 1.5 mg/L; plantlets from shoot apical meristem repressed with 6-BA at 2.0 mg/L; the most number of budding shoots in per explants reach 4. Isolated meristem shoot tips infected with Agrobacterium tumefacien have strong endurance on infected damnification and kan, of which selective concentration reach 100mg/L.
