Extraction Purification And Nature Research Of Earthworm Antibacterial Peptide

The antimicrobial activities of peptides from earthworm coelomic fluid extracted with electric shocks and E.Coli.induction+electric shocks were studied.Theose results show that:after 0h,3h,4h,5h,and 6h induction with E.Coli.,the inhibition diameter of E.Coli.were 19.7mm,18.00mm,15.33mm,17.33mm and 0mm respectively.The results also showed that inhibition diameter declined with the induction time.inhibition effect of Aspergillus flavus in diameter were 16.00 mm,14.00mm,13.00mm,12.00mm and 10.00mm respectively,the best was direct electric shocks.Inhibition diameter of Aspergillus flavus also reduced linearly with induction time with E.coli.The E.coli inhibition diameter wes larger than the reports(E.coli inhibition effect of 19.7mm:11.00mm,0mm,5.2～6mm),and found that the antimircobial peptides form earthworm were found that have anit-Aspergillus flavus effcts;There were no inhibitory effect of the peptides on Staphylococcus aureus, Pseudomonas aemginosa.After purified with Sephadex G-100 Chromatography,the Aspergillus flavus inhibitory effect was reducing,(antibacterial circle diameter of 16mm: 12mm);and there were no inhibitory effect on E.coli(antibacterial circle diameter of 19.7mm:0mm).The peptides molecular weight measured by SDS—PAGE were mainly in the 14.4～20.0 KDa,and one zonewas less than 14.4 Kda.So that the use of E.coli—induced and electric shocks to extract has anti-aflatoxin and the E.coli peptide, for the use of earthworms extraction of crude antimicrobial peptides provides a successful method and found the use of these methods of extracting the earthworm have a good the role of aflatoxin in order to resolve the aflatoxin contamination of feed for livestock and its hazards to provide a new trail through the chromatography separation and purification will lead to extract inhibitory effect of the decrease and disappear,the need to further improve its methods.
