Adiponectin gene was detected by PCR amplification and sequencing technology,the sequence of adiponectin gene was analyzed and the association with meat quality traits in pigs was evaluated.The results as followed:1.Two mutation loci ofadiponectin gene were validated,one was 180th A→G situated in exon 2 and the other was 1171st G→A situated in intron 2.The genotypes:AA,aa,Aa and dd,DD,Dd were in loci 180th and loci 1171 st respectively.2.There were two genotypes AA,dd in Jianhe xiang pigs,but genotypes AA,aa,Aa,dd,DD and Dd in hybrid pigs which showed polymorphism.The allele frequency of hybrid pigs were A: 0.3400,a:0.6600,D:0.7600 and d:0.2400 respectively.The distribution of genotype frequency in samples were AA:0.6364,aa:0.2364,Aa:0.1273,dd:0.6364,DD:0.3273 and Dd:0.0364 respectively.3.There were seven genotypes:AADD,aaDD,AAdd,aaDD,AaDD,AADd,AaDd in two variable sites of Adiponectin gene.The allele frequency were A:0.6182,a:0.0182,D:0.0727 and d:0.2909 respectively.The distribution of genotype frequencies in hybrid pigs were aaDD>AaDD>AAdd=Aadd>AADD=AADd=AaDd,and the allele frequency were d>A=D>a. But there was only one genotype AAdd in Jianhe xiang pigs.The distribution of genotype frequencies in samples was AAdd>aaDD>AaDD>Aadd>AADD=AADd=AaDd,and the allele frequency was A>d>D>a.4.Different genotypes in pig adiponectin gene and the association with meat quality traits were analyzed.The analysis indicated that the muscle tenderness and cooking percentage were significant difference between genotype AA and Aa(P<0.05).Cooking percentage in genotypes DD and Dd was significantly higher than that in genotype dd(P<0.01).Cooking percentage of genotype AAdd was lower than genotypes Aadd,aaDD,AaDD.But the cooking percentage of these genotypes was in the normal range.The muscle tenderness of genotype AAdd was significantly lower than genotype Aadd(P<0.05).Therefore,it was speculated that muscle tenderness was best,if the samples with homozygous genotypes AA,dd and AAdd.
