Immunogenicity And Application Of The HN Epitopes On Newclastle Disease Virus

NDV belongs to the paramyxovirus with a single envelope,the negative chain and no segments.It's genome is about 15 kb encoding six structural proteins.HN and F are the most important pathogen antigen and protective antigen of NDV,which play the most important role in the immune protection of ND Hemagglutinin-neuraminidase (HN) can adsorb corresponding cell receptors on susceptible cells,which recognize receptors in the course of virus infection,for it has the activities of hemagglutinin and neuraminidase.Epitope is also known as antigenic determinant is the antigen molecule antibody molecules can be identified sites in the protein antigen in general from several adjacent or directly adjacent conformation on the amino acid composition of the decision of the body to produce antibodies specific, humoral immunity induced by the basic unit.The humoral immune response to the main NDV,the identification of B-cell epitopes revealed ND is the essence of humoral immunity.In this study,based on two epitope peptide of HN.We have founded ELISA model,and assembled rapid strip.1,In the application of bioinformatics analysis of the chicken NDV HN protein structure characteristics on the basis of the overlapping peptide 18.After coupled with the protein BSA vector,check the reactivity of the synthetic peptide with monoclonal antibodies or chicken NDV by peptide ELISA and Dot-ELISA.The results showed that,the peptide has two linear epitopes which can be identified by monoclonal antibodies:HN02.DTLEKHTLRSETSTYN;HN03.ETSTYNLTVGDTGSGL;The epitope peptide identified by monoclonal antibodies can also react with chicken NDV antiserum.Chicken,immuned with HN02-BSA and HN03-BSA,can obtained the antibody for peptides,it's suggested that these epitopes on NDV be valuable in development of peptide-based vaccines for NDV.2,Based on two epitope peptide of HN,ELISA was founded,which sensitivity is the same as HI's,more,Immunochromatographic lateral flow test strip was primally study,limit of detection can reached 61g2 in 10 minutes.
