Clanis bilineata tsingtauica Mell, a kind of tranditional edible resource insects, is in Clanas, Sphingidae, Lepidoptera on entomortaxy. With the exploration of nutritive value of C. bilineata tsingtauica and people's increasing demanding on green food of insect, the market of larva C. bilineata tsingtauica enlivens gradually, resulting in the remarkable trends of industrialization of this insect. With the Clanis bilineata tsingtauica Mell's transfering from pest of legume crops to edible insect resources, which has resulted in special breed project of C. bilineata tsingtauica, and created considerable benefit, many local governments and enterprises pay much attention to it. Due to the popularity of C. bilineata tsingtauica, it has become the local characteristic cate in the surrounding area of south Shandong and Lianyungang.Previous study on C. bilineata tsingtauica mostly focused on the effect of environmental factor, such as perature, humidity, illumination and density ect. While, this paper will study the nuritional pysiology and its effect on host of C. bilineata tsingtauica from the point of view of microscology, in order to exploit the dominant intestinal strains of C. bilineata tsingtauica, changing the backward status of production depending on natural breeding and exploring a set of artificial breeding technique suitable for factory production of C. bilineata tsingtauic. Study on intestinal bacteria of first to fifth instar larva, overwintering larva and pupa of C. bilineata tsingtauica was conducted. The major research results are showed as follows.First, 23 intestinal strains isolating from first to fifth instar larva, overwintering larva were identified as Staphylococcus, B. lichenifomis, Brachybacterium, B. subtilis, C. pusillum, B. clausii, Cellulomonas, B. circulans, K. axytoca, S. lentus, Propionibacterium, Terrbacter, Proteus sp., Rahnella, E. cloacea, A. thermoaerophilus, Cedecea netei, Hafnia, Serratia, B. weihenstephanensis, B. pumilus, M. varians, P. gluconolyticus, 3 intestinal strains isolating from overwintering larva were identified as Staphylococcus, B. subtilis, Propionibacterium; Staphylococcus was isolated from pupa.Second, Staphylococcus and Bacillus were isolated from the overwintering larva from first to fifth instar larva, which proves that these two kinds of bacteria are resident ones in larvae C. bilineata tsingtauica. In the meantime, among all sample intestinal bacteria separated, Staphylococcus and Bacillus overpower others in quantity. From the two features of quantity and residence, Staphylococcus and Bacillus are dominant bacteria of larva C. bilineata tsingtauica.Third, from the physiological-biochemical tests on intestinal bacteria of the larva C. bilineata tsingtauica, it was found that most bacteria can exist at high temperature and be cultured on PDA medium. Meanwhile, they have strong capability in using sugar alcohol and decomposing macromodecule, which is corresponding to the natural occurance of larva C. bilineata tsingtauica in hot season from July to September.Fourth, the sequences of 16SrDNA were obtained from two dominant strains through further identifing. The result shows that they are highly homologous to Staphylococcus and Bacillus, it is coinsident with the appraisal result of colony morphology and physiological-biochemical tests.
