Clone And Analysis Of The 5' DNA Sequence Of Vitellogenin From Antheraea Pernyi

This thesis mainly contains the clone and analysis of DNA sequence of the 5' -flanking region of vitellogenin in one of economic insects with spinning and cocooning Antheraea pernyi,which belongs to Lepidoptera.There are little study related to the 5' -flanking region of vitellogenin gene at home and abroad.In this paper,two primers were designed based on the 5' -flanking region of ApVg.An about 500bp DNA fragment is obtained through PCR from Antheraea pemyi genomics DNA.The 5' -flanking region gene sequences of ApVg is also obtained through cloning,sequencing and analysis.The transcription initiation site of ApVg gene is 55 bp upstream from the translation start codon(ATG).The result shows TATA box is present at -79～-86bp from the translation start codon(ATG).A.B.mori doublesex homologue(BmDSX) binding site(ACATTGT) was found at -156～-163bp from ATG.Elements derived from other insents,such as CREB,GATA,and CdxA,are also found in the 5' -flanking region.Compared the 5' -flanking region of ApVg with Bombyx mori's and Antheraea yamamai's,We can seen that the 5' -flanking region of ApVg has many similarity with Antheraea yamamai's.But the similarity the 5' -flanking region of ApVg with Bombyx mori's is less than it with Antheraea yamamai's.The shows that the homology of Antheraea pemyi and Antheraea yamamai is higher than it of Antheraea pemyi and Bombyx moil.The analysis of the 5' -flanking region sequence of AyVg can help us for the study of the function of the 5' -flanking region sequence of AyVg and show that the DSX is a possible sex determination gene.This sequence has been submitted to GeneBank,the accession number is EU330205.
