| Bermudagrass[Cynodon dactylon(L.) Pers]is a widespread species in the world,and widely distributed in the Yellow fiver basin and its south regions and Xinjiang province,it is popular with the characteristic of dwarfness,fineness,high capacity to propagate, resistance to high temperature,drought and trampling,the fast turf formation speed.At present,most of turf bermudagrass used in China are the Tif series hybrid bermudagrass from the United States and commonly use vegetative to establish lawn.There are kinds of methods in vegetative establishment,including paving turf and spreading stem etc.Paving turf is the common way,but this approach seriously destroys cultivated horizon,resulting in bogging down of the arable land,acceleration of soil salinization,pollution of the environment,and damage the balance of ecology.Spreading stem is the best way of vegetative establishment by the comprehensive comparison of formation speed,quality and homogeneity recently.In addition,soiless sod and plant strip for turf establishment are also ecological and environmental.All of above related to the question of post-harvest stolon machining and storage.The isolated stolon's metabolic activities are continuing.Therefore, the questions of how to maintain the water content and delay aging,prolonging the storage period are anxious to solve in practice.The variation of apparent growth indexes and physiological and biochemical indexes of the stolon in saturated moisture content were studied at high temperature 35℃(day)/ 27℃(night),normal temperature 25℃(day)/17℃(night) and low temperature 15℃(day) / 7℃(night) for two weeks,respectively.The apparent growth indexes included survival rate,tiller number,biomass,etc.The physiological and biochemical indexes included respiration rate,sugar content,and so on.Finally,the reasonable concentration of three kinds of growth regulators(SA,TE,PP333) was studied.According to the results of screening,the mechanism of delaying the aging was discussed.The main results were showed as follows: 1 Effects of different storage temperature on regeneration energy of isolated hybrid bermudagrass stolonHigh temperature 35℃(day) / 27℃(night) has great influent on regeneration energy of isolated stolon in early storage.The survival potential and survival rate declined sharply and stolon all died after two weeks.But under low temperature,the survival potential of stolon was still 85.0 percent after two weeks.The effects of normal temperature and low temperature on tiller number,biomass and other apparent growth indexes were small.There was significantly positive correlation among indexes,so all indexes can reflect the impact of different storage temperature on the stolon's regeneration energy.2 Effects of different storage temperature on physiological and biochemical indexes of hybrid bermudagrassTwo obvious respiratory peaks were appeared under high temperature,and the respiration rate was markedly higher than that under normal temperature and low temperature(P<0.05).Under high storage temperature,high respiration consumed large amount of soluble carbohydrate,which lead to the fastest decline speed and biggest decline scope of soluble sugar content,the relative conductivity increased by 51.55%.Respiration rate has greatly significantly positive correlation with relative conductivity,has greatly significantly negative correlation with sugar content;and relative conductivity has greatly positive significant correlation with sugar content.3 Effect of different plant growth regulator on survival rate and relative conductivity of hybrid bermudagrass isolated stolonThis section used different concentrations of three kinds of plant growth regulators (SA,TE,and PP333) to treat with potted hybrid bermudagrass.The survival rate and relative conductivity of stolon were studied after the stolon storied at high temperature 35℃(day)/27℃(night) for one week.The results showed that the salicylic acid 1.0 mmol/L was the reasonable concentration.Under this concentration,the survival potential of stolon was highest,reached to 73.3 percent;and the relative conductivity was 42.8 percent.Both were remarkable different than controlâ… (P<0.05 ) However,under the treatment of trinexapac-ethtl and paclobutrazol with different concentration,the survival potential and relative conductivity of stolon were not remarkable different than controlâ…¡.4 The preservation research of reasonable concentration salicylic acid on isolated hybrid bermudagrass stolonThe reasonable concentration of salicylic acid can restrain the water loss of stolon. Under control treatment,the stolon reached 50 percent lethal at the 5th day.Under salicylic acid treatment,the stolon reached 50 percent lethal at the 8th day.Early in storage,the rate of respiratory was declined with salicylic acid treatment.The content of chlorophyll and soluble protein decreased slowly.The activity of SOD was improved.To a certain extent, salicylic acid reduced the damage of the cell membrane system,delayed the aging of the isolated stolon. |
