Sclerotinia sclerotiorum belongs to Ascomycotina, Helotiales, Sclerotinia, a worldwide plant pathogen which could parasitize in more than 400 different host plant species.Due to the lack of the natural environment can be used in the practice of Sclerotinia resistance germplasm resources, host of Sclerotinia sclerotiorum resistance is controlled by multiple genes and genetic mechanisms for interaction between the complex. Practice has proved that the disease occurred after the production of chemical control is the only effective control measures. Disease is mainly spread of Sclerotinia sclerotiorum ascospores in the sexual stage of a disk wind Ascospore release of the spread of disease to increase the spread of Sclerotinia sclerotiorum. The study is particularly important phase of sexual reproduction.The main role of the mating types, common to all studied ascomycetes, is to control the recognition mechanism leading to fertilization and the evolution. Each of the mating type idiomorphs (MAT-1 and MAT-2) consists of approximately 1.2 kb of unique DNA flanked by sequences that are almost identical between chromosomes, which are alpha-box and HMG-box, conserve sequence in mating gene. In this study, we constructed phylogenetic tree of 227 species of Ascomycotina fungi baseing on alpha-box and HMG-box sequence and do analysis about the relationship of evolution between them which were in high conformity. Deep Hypha phylogenetic tree focused on the same major loci, including the well characterized nuclear rRNA genes, and the protein-coding loci tef1 and rpb1 and rpb2.The analysis results are as same as the front.This article focuses on Sclerotinia sclerotiorum MAT mating type genes have been studied for the genetic analysis of Sclerotinia sclerotiorum work laid the foundation. In this study, the left and right select the MAT gene flanking sequence of about 1000bp of the knockout vector sequence of homologous recombination, using PCR technology around flanking sequence, the correct sequence, respectively, after the reorganization of the Agrobacterium-mediated transformation vector pBI-G3CN to construct by Agrobacterium-mediated transformation of Sclerotinia sclerotiorum knockout vector ofâ–³pBI-G3CN-MAT, and transformed to Agrobacterium tumefaciens EHA105 cells shocks in.In short, this study has been successfully cloned Sclerotinia sclerotiorum MAT mating type gene sequences flanking the left and right, build the MAT mating type gene knockout vector ofâ–³pBI-G3CN-MAT. The first use of Sclerotinia sclerotiorum ascospores establish Agrobacterium-mediated genetic Sclerotinia sclerotiorum into the system, and optimize the system conditions for the development of new pesticides and laid the foundation for disease prevention and control.
