Eriocheir sinensis(commonly called river crab) is one of the developing and economically beneficial agricultural products in China.Bacteria related diseases would inevitably occur as a result of high density cultivation,therefore,the use of antibiotics is a general practice in the E.sinensis cultivation.To ensure the safety of food from animal origin,the residues of enrofloxacin(ENR) and its metabolite ciprofloxacin(CIP) should be studied.The objectives of the present study are to provide information on the kinetic of ENR and CIP in E.sinensis at different water temperature,administration regimes and dose and gender,The results were as follows:1.A pharmacokinetic study of ENR following a single intramuscular administration(3.0 mg·kg-1) was carried out in the Chinese mitten-handed crab at 17,23 and 29℃water temperatures.Hemolymph,muscular and hepatopancreas samples were collected at 1,10 min and 0.5,1,2,8,16,24,48,72 and 168 h post-injection.The tissue level of ENR and CIP were simultaneously determined by high-performance liquid chromatography method. The ENR level in hemolymph and muscle reached maximum momentarily,while those in hepatopancreas appeared to be a maximum then declined.The data was analyzed with Practical Pharmacokinetics Program(3P97).The kinetic profile of ENR in hepatopancreas was conformed to a two-compartment model.At 17,23 and 29℃,the absorption half-life (1.81,0.97 and 0.14 h,respectively),elimination half-life(240.27,60.57 and 37.59 h, respectively) and the area under the concentration-time curve from zero to infinity(2148.65, 324.16 and 264.47μg·mL-1·h-1,respectively) of ENR in hepatopancreas was found to decline with temperature rised.Whereas the total body clearance rise with temperature,and which were 0.0014,0.0093 and 0.011 L·kg-1·h-1,respectively.2.A pharmacokinetic study on ENR following different administration regimes(i.e. intramuscular,oral and bath administrations) was carried out for the Chinese mitten-handed crab(Eriocheir sinensis) at 17℃water temperature.Muscle and hepatopancreas samples were collected at each sampling time of 1,10 min,0.5,1,2,8,16,24,48,72,168 h for intramuscular administration,0.25,0.5,1,2,4,6,8,12,24,48,72,168 h for oral and bath administration.The levels of ENR and CIP were simultaneously determined(n=5) by high-performance liquid chromatography.The results showed that similar absorption and elimination patterns of ENR level in muscle and hepatopancreas were observed for oral administration and bath treatment,however,the trend in ENR level changes in muscle and hepatopancreas after intramuscular administration was quite different from oral and bath treatments.Meanwhile,the level of ENR in hepatopancreas was much higher than that in muscle for the same administration regimes.The concentration-time data was analyzed with Practical Pharmacokinetics Program(3P97),the kinetic profiles of ENR in hepatopancreas and muscle were conformed to two-compartment models.The absorption and elimination of ENR in hepatopancreas for bath treatment was faster than those for oral and intramuscular administrations.However,the pattern was quite different in muscle,the fastest absorption and elimination of ENR in muscle was seen for intramuscular administration.3.A pharmacokinetic study of ENR at different administration doses(6 and 12 mg·kg-1) and sex was carried out in the Chinese mitten-handed crab at 25℃water temperature. Hemolymph,muscle and hepatopancreas samples were collected at each sampling time. The tissue level of ENR was determined by high-performance liquid chromatography method.The results showed that similar absorption and elimination pattern that ENR level declined fastly then slowly were reached in ENR level in hemolymph at different administration dose.Sirriilarities were found among the hemolymph,muscle and hepatopancreas concentration-time curve at both sex,and ENR level peaked within 1 h post-administration followed by a second absorption peak at 8 h post-administration in female and 16 h post-administration in male.The kinetic profiles of ENR in hepatopancreas and muscle were conformed to two-compartment models.The result showed that ENR in hemolymph of Eriocheir sinensis absorbed and distributed obviously faster at 12 mg·kg-1 than 6 mg·kg-1,and the female Eriocheir sinensis absorbed and distributed obviously faster than the male.
