In this study,effects of Genistein and SDG on the mammary development and related sex hormones in female adolescent rats were studied separately by gastric infusing and dietary Genistein and SDG into basal diets.The growth of mammary gland was evaluated by RP-HPLC,RIA,tissue slice,RNA/DNA,et al.On the base of these,the mechanism of these two phytoestrogens Genistein and SDG on mammary development and related sex hormones in female rats were discussed.This research includes three series:Series 1:Effects of Genistein on mammary development and related sex hormones in female adolescent ratsEffects of Genistein on the mammary development and related hormones in female adolescent rats were studied separately by gastric infusing and dietary Genistein to evaluate the impact of Genistein on the development of mamogenesis and its possible mechanism..The indexes were evaluated by RIA,tissue slice,RNA/DNA,et al.During the results by gastric infusing Genistein,the group of Exp.mammary gland relative body weight was significantly increased for 35.41%(P<0.01);and the contents of RNA and RNA/DNA in mammary tissue were separately significantly increased for 27.24%and 22.07%(P<0.01);The result of RIA showed that the serum 17β-E2 and progesterone levels in the group which was administered with Genistein were separately significantly increased for 26.68%and 130.54%(P<0.01).During the results by dietary Genistein, mammary gland relative body weight was significantly increased for 25.45%(P<0.01); and the contents of RNA and DNA,RNA/DNA in mammary tissue was separately significantly increased for 24.49%(P<0.01),10.54%(P<0.05) and 12.63%(P<0.05);The result of RIA showed that the serum 17β-E2 and progesterone levels were separately significantly increased for 9.95%(P<0.05)and 247.92%(P<0.01).Conclusion: Genistein could promote the development of mammary gland of female adolescent rat. Maybe vin neuroendocrine pathway,and the results of gastric infusing and dietary Genistein were nearly the same. Series 2:Effects of SDG on mammary development and related sex hormones in female adolescent ratsEffects of SDG on the mammary development and related hormones in female adolescent rats were studied separately by gastric infusing and dietary SDG to evaluate the impact of flaxseed lignans and its main metabolites on the development of mamogenesis and its possible mechanism.The indexes were evaluated by RIA,tissue slice,RNA/DNA, et al.During the results by gastric infusing SDG,the group of Exp.mammary gland relative body weight was significantly increased for 23.11%(P<0.01);and the contents of RNA and RNA/DNA in mammary tissue were separately significantly increased for 26.34%and 23.11%(P<0.01);The result of RIA showed that 17β-E2 and progesterone levels in the serum in the group of experiment which was administered with SDG both were separately significantly increased compared with the group of control(P<0.01).During the results by dietary SDG:SDG enhanced mammary gland growth;increased ductal and alveolar volume;the concentrations of DNA,RNA and RNA/DNA were significantly increased in mammary tissue.Compared with the group of control,the serum 17β-E2 and progesterone levels in the group of ExperimentⅠwhich was supplied with SDG both were significantly increased,while only progesterone levels was significantly increased compared with group of ExperimentⅡ(P<0.01).Conclusion:Flaxseed lignans and its two main metabolites could promoted the development of mammary gland.and may be vin direct action and neuroendocrine pathway and the results of gastric infusing and dietary SDG were nearly the same.Series 3:Determination of Lignans in Serum and Mammary Gland Tissue of Rats by Reverser-phase High Performance Liquid ChromatographyThe flaxseed lignans,as one of the main phytoestrogens,could be converted to mammalian lignans by the intestinal flora present in humans and rats.Both mammalian and plant lignans have been shown to possess weakly estrogen and antiestrogen activities.The lignans concentrations in serum and mammary gland of rats were measured by Reversed-phase high performance liquid chromatograph(RP-HPLC).The result of gastric infusing SDG disclosed that,both SDG and mammalian lignans(mainly enterolactone and enterodiol) concentrations were significantly increased in serum and mammary gland tissue which compared with the group of control(P<0.01);Also,the result of dietary SDG showed that,both SDG and mammalian lignans concentrations were significantly increased in serum.and mammalian lignans in mammary gland tissue were also significantly increased which compared with the group of control(P<0.01),while the experimentalⅡ,which group was supplied with flaxseed lignans+antibiotics simultaneously,both SDG and END,ENL in serum and mammary gland tissue were significantly decreased(P<0.01) compared with the experimentalⅠthat was fed with flaxseed lignans.Conclusion:Flaxseed lignans and its two main metabolites could be absorbed into serum even mammary gland tissue,and may be vin direct action and neuroendocrine pathway to promote the development of mammary gland.
