Selenium and zinc is an essential trace element for animals and human.It has been found that they had some important biological functions;functions of selenium include improving immunity,preventing cancer,resisting flee radicals,postponing aging, detoxification from some heavy metals,and preventing some local epidemic etc.Because of the important physiological and biochemical functions,Zinc go by the name of life element.Functions of zinc include promoting growth,improving immunity,enhancing reproductive performance,participating in metabolism of protein and nucleic acid, regulating activity of enzyme etc.Nowadays,It has caused attention by microbiology enriching trace element to attain organic and low cost additive of trace element.Comparaed with inorganic selenium and inorganic zinc,organic selenium and organic zinc have advantage of higher bioavailiability,low toxicity and less potential environment pollution. Yeast has not only competence of enriching trace elements,is but also one of germ which is fermented in industry.Other germs had a competence of enriching many trace elements(eg, lactobacillus).Product of organic trace element which is used to human and animal come out.The study had produced a selenium-enriched and zinc-enriched probiotics which combines the virtues ofprobiotics with the virtues of organic selenium and organic zinc by screening probiotics strains and optimizing culture condition of selenium-enriched and zinc-enriched probiotics,and its effect was studied1.The Screen of probiotics strain enriched selenium and zinc.In this experiment,3 Yeasts and 5 Lactobacillus were screened by Se-tolerant and zinc-tolerant experiments.Se-tolerant experiment were done by inoculating Yeasts and Lactobacillus into agar plate culture added different concentration Se4+,zinc-tolerant experiment were done by inoculating Yeasts and Lactobacillus into agar plate culture added different concentration Zn2+,High Zn-tolerant and Se-tolerant strainsY2 and LA were screened synthetically. 2.Culture Conditions Optimization of Selenium-enriched and zinc-enriched probiotics.Using Candida utilis and Lactobacillus acidophilus as probiotics strains,adding inorganic Zn2+ and Se4+ into culture medium.Selenium-enriched and zinc-enriched probiotics was producted by mixed cultivation,and Optimizing culture conditions of Selenium-enriched and zinc-enriched probiotics,by screening,The optimum culture conditions for selenium-enriched and zinc-enriched probiotics were that,using special culture medium,adding inorganic-selenium concentration of 5μg·ml-1 at the medium, adding inorganic-zinc concentration of 150μg·ml-1 at the medium,Medium of 50ml in a 250ml flask.Inoculation volume:5%,cultivated temperature:32℃,cultivated time:36h. Under the culture conditions,selenium-enriched and zinc-enriched probioties were mixedly cultured,cultivated liquid was centrifugated in low temperature,added cryoprotectants into mud of probiotics,vacuum freeze-drying.Product of selenium-enriched and zinc-enriched probiotics was attained.Mass of probiotics products was 25.94g·L-1,the content of selenium in the product were 167.23μg·g-1 the content of zinc in the product were 4.52mg·g-1,the colony forming units of candida utilis in the product were 1.02×108 cfu·g-1, the colony forming units of Lactobacillus acidophilus in the product were 1.48×109cfu·g-1.3.Effects of selenium-enriched and zinc-enriched probiotics on content of selenium and zinc in blood,antioxidative activities and intestinal microflora in canines.To investigate the effects of selenium-enriched and zinc-enriched probiotics on the content of selenium and zinc in blood,antioxidation function and intestinal microflora in canine.Eight 18-month native canines,female and male in half,were randomly divided. into the control and treatment groups on average.The control canine received base diet throughout the experiment,the treatment group received basic diet supplemented with 2.0 g selenium-enriched and zinc-enriched probiotics everyday.To determine experimental indexes,the samples were collected on day 0,on dayl5 and day 30,respectively.Results: Compared with the control group,On day 0,the indexes that were content of selenium and zinc in blood,blood GPX,serum SOD,T-AOC and the amount of Lactobacillus in the excrement were significantly increased,with the amount of Escherichia coli in the excrement significantly decreased,while the indexes that is serum MDA and the amount of Bifidobacterium,Staphylococcus and Enterococcus in the excrement have no significant change;On days 30,the indexes that were content of selenium in blood,serum SOD,serum T-AOC and the amount of Lactobacillus in the excrement were very significantly increased, with content of zinc in blood,blood GPX and the amount of Bifidobacterium in the excrement significantly increased;the amount of Escherichia coli in the excrement was very significantly decreased,with the indexes that were serum MDA,the amount of Staphylococcus and Enterococcus in the excrement very significantly decreased. Conclusion:Selenium-enriched and zinc-enriched Probiotics could increase content of selenium and zinc in blood and antioxidation function,improve and regulate the intestinal microflora.
