The germplasm resource of Pleurotus(Fr.) Kumm is abundant. There are more than 100 kinds of Pleurotus(Fr.) Kumm which discovered over the world at present, 36 kinds were discovered by our country . In long-term cultivation domestication process, For its shape characteristic is extremely easy to be effected by the external environment condition, the same species have large differences in it's phenotype characteristic for different growth environment, then cause the phenomenon of homologous synonym and dissimilar homonymous. AFLP is usually used for carry on the comparison between different species or during the different strains in inter-species by it's high technical sensitivity, well repeatability, high resolution. In this paper, diversities, classification, genetic relationship and species identification between 30 kinds of Pleurotus(Fr) Kumm cultivars were evaluated and analyzed by AFLP, which Provide theoretical foundation for the evaluation, classification, selection of high quality rermplasm resources, protection and utilization of Pleurotus(Fr.) Kumm. The results indicate that:1. Mycelium genome team DNA which directly cultured in PDA medium was be extracted by using CTAB+ free washing method. The result indicated that, the DNA relative molecular mass close to 21kb, EcoRI holoenzyme cuts, and also can obtained the clear PCR atlas and the AFLP fingerprint atlas.2. This research selects 9 pairs direct in the combination. And a total of 1830 bands were amplified ,1799 are the polymorphism mark, the polymorphism average percentage is 98.3%, this test indicated the result of every primer combination's expanding show the rich polymorphism between different materials . The similar coefficient scope between used material is 0.37~0.89, between P777 and P999 (0.89) is the highest, The genetic relationship between them are relatively close. The lowest is between Yuxiu3hao and 615 (0.37).The genetic relationship between them are relatively far. 3. 30 kinds of cultivars can be divided into three groups according to the combine line (0.51) of the genetic distance .According to the combine line (0.62)of the genetic distance ,30 kinds of cultivars can be divided into four groups This result provide theoretical basis of the classification of Pleurotus(Fr.) Kumm.4. 30 kinds of germplasm resources of Pleurotus(Fr) Kumm were used to be identificated by AFLP. The result showed that 29 kinds of cultivars have AFLP fingerprinting in the 30 kinds of Pleurotus(Fr.) Kumm Plants, the number of AFLP fingerprinting most reached as high as 15, but only one special belt's existence or the flaw may differentiate with other resources in identification of a species .It fully show that AFLP technique is a powerful and efficient tool in identification of Germplasm resources.
