The Identification, Cloning And Phylogenetic Evolution Of A Y-Type HMW-GS From Triticum Tauschii

Triticum tauschii(Aegilops tauschii,2n=2x=14,DD),which is belonging to Triticum tauschii,is common wheat D genome donor.The existence of a wide range of Triticum tauschii HMW-GS Allozyme variation,its genetic variability as compared with ordinary types of the wheat genome enriched coding.Isolation and Cloning of these genes in wheat breeding for a better position to offer a wealth of genetic resources for the study of the origin of hexaploid wheat and evolution has provided useful information on common wheat used in mining for quality improvement of new genetic resources of great significance and application prospects.In this study,the SDS-PAGE technology and AS-PCR technology for polymer Triticum tauschii glutenin subunits were analyzed and identified,And to gene amplification,cloning and sequencing, and conducted a phylogenetic solution analysis.The main findings are as follows:To being SDS-PAGE analysis for 68 Triticum tauschii,It was found nine kinds of x-subunit, five kinds of y-subunit,33 kinds of combination of types,showing the diversity of portfolio, Triticum tauschii showed that the composition of high molecular glutenin polymorphism.Triticum tauschii from the 67 discovered a new type of polymer 1 kinds of glutenin subunits Ty＜12,was named Dy8.1~t.Have identified a new subunit of the special tauschii and Chinese Spring genome amplification of AS-PCR.Triticum tauschii through the new subunit gene was amplified by x-subunit and the y-subunit,recovery y-subunit,as a cloned fragment.Cloning and sequencing of the Triticum tauschii Dy8.1~t subunit(GenBank accession number FJ499503),Its full-length coding region of 1995 nucleotides,which has the deduced amino acid sequence of y-type high molecular glutenin subunits of the typical characteristics.The sequence contains 23 six-peptide(PGQGQQ) and 4 nine-peptide (GYYPTSLQQ),the subunit encoded by the D genome of the y-type glutenin subunits in the polymer repeat one of the shortest.Cloning and sequencing of the Chinese Spring By8~* subunit(GenBank accession number GQ121361),Its full-length coding region of 1980 nucleotides.Its deduced amino acid sequence with a y-Type high molecular glutenin subunits of the typical characteristics,the sequence containing 20 six-peptide(PGQGQQ) and 4 nine-peptide(GYYPTSLQQ).Have been cloned from Triticum tauschii Dy8.1~t polymer obtained glutenin subunits and D genomes of wheat and wheat B genome sequence at the molecular level have a greater difference; From Chinese Spring has been cloned By8~* Polymeric glutenin subunits from Triticum tauschii and the D genome sequence at the molecular level there are large differences.Of Triticum tauschii glutenin subunit polymer research for wheat quality improvement experiments and lay a firm theoretical basis.
