The obscure puffer Takifugu fasciatus is a related species of the Genome Model species Takifugu rubripes.However,the mechanism and process of the respond to chemical stimulus in them has not been found up to now.We know that there is a gene family named taste receptor which is related to the chemical sensation in the Takifugu rubripes and Danio rerio,however no research about how to explain relationship has been reported.This paper which depends on the sensory behavior by means of the statistics under some different chemical stimulus was in order to obtain the regularity of the sensory behavior.The regularity shows among the animal muscles the samples love the chicken; among the plants the samples love celery;among the amino acid like derivant they love glycine,alacine,taurine and DMPT.There is an avoidance response from yeast paste. Another behavior phenomenon imply the samples hardly love nucleotideThe sequences of Taste receptor gene of some fish were published on NCBI.PCR and RT-PCR are used to study the existence and the expression of Taste receptor gene in different tissues to discuss the function of taste receptor in Takifugu fasciatus.Taste receptor candidate gene T2R1 are differently expressed in different tissues.It is not expressed in thymus gland,head kidney and tongue,but expressed in oral epithelium,and is multi times expressed in normality.The results showed that Taste receptor gene is not possibly the key gene to response to chemical stimulus,however,the results probably can provide clues to find the true key gene about chemosensory behaviorThree DNA fragments including ORF of Takifugu fasciatus taste receptor gene were obtained.The amplified fragments named T2R1 are 921 bp,which only has 7 bases (Speculated fusion Protein 5 amine acid) different from that of Takifugu rubripes. Sequence homologous is 99.2%.The amplified fragments named T1R1 are 3376bp,which only has 27 bases(Speculated fusion Protein 11 amine acid) different from Takifugu rubripes.Sequence homologous is 98.9%.The amplified fragments named T1R2b are 312 bp,which only has 19 bases(Speculated fusion Protein 8 amine acid) different from Takifugu rubripes.Sequence homologous is 99.2%.In addition,two DNA fragments related T1R2a also are obtained.Sequence homologous was 98.3%.Bioinformatics analysis display that T1R1 and T1R2b which contained 6 extrons and 5 introns are consistent with the structure of the T1R gene family;that T2R1 without intron is Consistent with the structure of the T2R gene family.Speculated fusion protein contained the structural domain of the T1R gene family;These results can provide the evidence that we obtained the real taste receptor from Takifugu fasciatus and the clues to study the relationship between the taste receptor and chemosensory behavior.
