The disease exudative epidermitis (EE) in pigs is caused by infection with Staphylococcus hyicus. Studies have shown that both virulent and avirulent S.hyicus can be isolated from diseased pigs. The virulent strains produce an approximately 27-kD or 30-kD exfoliative toxin. S. hyicus exfoliative toxin has been shown to be the factor responsible for the alterations of the skin in EE in pigsThe ExhB gene, ExhB 5' gene and ExhB 3' gene, which had been cloned into a prokaryotic expression vector pET32a, were transformed into E.coli BL21(DE3) and induced by IPTG. After soluble analysis the fusion protein was furified. The culture was collected and tested by SDS-PAGE and indirect ELISA. Three pieces of protein were covered on ELISA plate separately, and done the cross-over experiment with anti-serum. The result shows ExhB1 had higher immunogenicity than ExhB2 in the experiment of ExhB anti-serum. It is possible that the antigen epitopes of ExhB is at the 5'end of ExhB gene.
