The Cloning And Sequence Analysis Of Hoxa11, Hoxd11 And Hoxc8 Gene Of Mongolian Sheep

We have successfully cloned 1673bp of Hoxc8 gene of sheep for the first time including all part of the Intron and the majority part of the Exon on this research,applying PCR methods and using 38 Uzhumchin sheep and 22 Sunid sheep as subjects.In the mean time,after an alignment research on Uzhumchin sheep and Sunid sheep it had been found out 7 differences between Uzhumchin sheep and Sunid sheep:(1) the 373^rd of the Hoxc8 gene series of Sunid sheep is C,while the one of Uzhumchin sheep mutated into T;(2) the 576^th of the Hoxc8 gene series of Sunid sheep is T while the one of Uzhumchin sheep mutated into A;(3.4) the 655^th and 656^th of the Hoxc8 gene series of Sunid sheep are C and G,while the ones of Uzhumchin sheep mutated into T and A;(5) the 727^th of the Hoxc8 gene series of Sunid sheep misses A;(6) the 1031^st of the Hoxc8 gene series of Sunid sheep is A while the one of Uzhumchin sheep mutated into T;(7) the 1566^th of the Hoxc8 gene series of Sunid sheep is T while the one of Uzhumchin sheep mutated into C.At the same time Hoxa11,Hoxd11 and Hoxc8 gene's nucleotide sequence of the Exon region for the Uzhumchin sheep and Sonid sheep also aligned in this experiment.The results show that the ratio of:the Hoxa11 gene was amplified by the second Exon fragment size of 192bp,Uzhumchin sheep,there is a deletion of a nucleotide and two mutation of two nucleotide;the Hoxd11 gene was amplified by the second Exon of 151bp fragment size(accessionGQ300854),Uzhumchin sheep,there is a mutation of one nucleotide; Hoxc8 gene was amplified by the first Exon of the size of 327bp fragment,there was no difference between Uzhumchin sheep and Sunid sheep.
