| The grading up dosage of yeast culture(YC) and disodium Fumarate was selected in vitro batch culture.Six guanzhong lactating goats with permanent ruminal cannulas were Induced SARA by feeding concentrate daily in gradually increasing feeding regime. The experimental animals were fed both yeast culture (YC) and disodium fumarate into diets when animals were in SARA period.The ruminal fermentation parameters were determined and the amounts of bactaria were quantitated by the traditional roll-tube technique and 16S rRNA oligonecleotoide probes hybridization. The results of this research as followed:(1) The result of vitro batch culture indicated: the pH and the cumulative gas production of nine combinations increased significantly compared with the control (P<0.05),while the NH3-N concentration of nine combinations decreased significantly (P<0.05). As compared with the control,total volatile fatty acids (TVFA) and propionate concentration increased significantly (P<0.05),while acetate/propionate ratio and acetate concentration decreased significantly (P<0.05).(2) The MFAEI of seventh combination was hightest(0.2842) by assessment of pH,cumulative gas production and NH3-N concentration. When respective addition of YC and disodium fumarate are 0.09g and 0.006g,the effect of fermentation is the best bycomparing the MFAEI.(3) As compared with the control, The grading up combination made the pH value,acetate,butyrate and TVFA concentrations increase significantly (P<0.05),while propionate concentrations and acetate/propionate ratio were not affected.The NH3-N concentration decreased significantly(P<0.05).(4) As compared with the control,The grading up combination made the populations of Amylolytic bacteria,S.ruminantium,M. elsdeni and the three kinds of predominant cellulose-decomposing bacteria increase significantly (P<0.05),while Lactobacillus and S.bovis decreased significantly(P<0.05). |
