| This experiment was conducted to investigate the appropriate ratio of 2-methylbutyrate, Isobutyrate, Isovalerate and Valeric acid in isoacids, and its appropriate add level in ruminant dietary. The effects of isoacids on gas production(GP), the bacteria nitrogen and the degradation of neutral detergent fiber(NDF) were studied by applying the artificial rumen technique. The milk cow performance and urine purine were analysed by add isoacids in the dietary in order to investigated the effect of isoacids.In this experiment, the results were shown as follows:Exp.1 The effect of four monomer isoacids on Goat Ruminal Fermentation in Vitro Gas ProductionUsed a single factorial trial, in accordance with the goat metabolic body weight, dry matter intake and dry matter content in the diet to determine the isoacids add level in the dietary is 0, 0.3%, 0.6%, 0.9%, and 1.2%, each level included 4 replicate. In vitro gas production method was conducted to study the effect of four monomer isoacids on goat rumen fermentation, by measuring the volume of gas production(GP), the bacteria nitrogen and the degradation of neutral detergent fiber(NDF), to select the appropriate proportion of added.According in vitro gas production, the appropriate levels of 2-methylbutyrate, Isobutyrate, Isovalerate and Valeric acid were supplemented 0.3%~0.6%, 0.6%~0.9%, 0.6% and 0~0.3%, respectively.And the appropriate ratio of 2-methylbutyrate, Isobutyrate, Isovalerate, Valeric acid in isoacids is 2:3:2:1(that is 25%, 37.5%, 25%, 12.5%).Exp.2 The effect of the compound isoacids on Goat Ruminal Fermentation in Vitro Gas Production Used A single factorial trial,in accordance with the goat metabolic body weight, dry matter intake and dry matter content in the diet to determine the isoacids add level in the dietary is 0, 0.1%, 0.2%, 0.4%, 0.6%,and each level included 4 repetitions.in vitro gas production method was conducted to study the effect of compound isoacids on goat rumen fermentation,by measuring the volume of gas production(GP), the bacteria nitrogen and the degradation of neutral detergent fiber(NDF),to select the most appropriate level of added.The results demonstrated,under in vitro gas production testing conditions,the appropriate levels of compound isoacids add to dietary is 0.2%~0.4%, and from the cost of economic efficiency considerations, chose 0.3% add level for the final selection in dairy cow diets.Exp.3 Isoacids additives on the performance of dairy cowsSelect 40 nutrition middle, health Chinese holstein Dairy cow, with similary of age, weight, fetal, and the period of lactation, were regional divided into control and trial groups, each with 20 cows. Both of two groups were fed the same basal ration, but cows in trial group were fed rations supplemented with isoacids additive at 51g/day per cow, and the control group were fed rations without additive. During the experiment, milk yield of cows in trial and control group was checked, samples of the mixed milk were tested milk quality and calculate cost- effectiveness.The results showed that the milk yield of the trial group is 1.45kg/d per cow higher the control's, increase by 8.06% (P> 0.05), equivalent to 3.34 yuan for the economic benefits, and the input-output ratio: 1:3.61; Compared with the control group, Fat, protein, lactose, the total dry matter, non-fat milk solids and somatic cells have a certain improvements, but the difference was not significant (P> 0.05).Exp.4 Effect of isoacids addiditive on purine derivatives of urine in milk cowIn the beginning of the 3 days, five milk cows were selected randomly from the control group and trial group to collect urine for 3 days. To avoid the urine purine derivatives to be damage and the ammonia volatile by bacteria, used the acidity machine to adjust the final pH value is below 3, in accordance with per day the proportion of urine in 3d, to make a 450ml mixed samples,kept at -20℃, use colorimetry to determine the allantoin content,and use the uric acid test kit to measure uric acid content.The results showed that add isoacids additives in the ration, the content of allantoin in the trial group has an increase of 4.41% (P <0.05) than the control group; the uric acid levels increased 0.70% (P> 0.05), but the difference was not significant. Urine purine derivatives in the trial group has an increase of 4.11% (P <0.05) than the control group. As the content of urine purinr derivatives and the rumen microbial protein synthesis highly relevant, it denotes that add isoacids additives in the ration can increase the milk cow rumen microbial protein synthesis. |
