Lily and Narcissus are important bulb flowers. However, It is extremely vulnerable against virus disease, the virus will cause a recession of its species and the loss of value of commodity production. Among them, Lily symptomless virus (LSV) occurred more commonly. LSV general will not cause obvious symptoms, but with the cucumber mosaic virus (CMV) or Lily mottle virus (LMoV) composite infection can cause transparent veins, mosaic, malformation, and the whole necrotic spot dwarfing symptoms. It is the key issues that the establishment of rapid, accurate and efficient virus detection system of virus-free Lily, Narcissus and the resumption of production of the ball.The laboratory has established multiple RT-PCR detection methods of three common viruses of Lily at the same time and the combination of stripping tip with HIV inhibitors virus removal methods. On this basis, this paper carried out the further research about lilies, narcissus virus detection and Lily symptomless virus (LSV) of virus-free technology.1. LMoV was detected in Narcissus pseudonarcissus from the Netherlands using one set of specific primer by reverse transcription-polymerase chain reaction (RT-PCR). The primer was designed according to the published sequence of the coat protein of LMoV in the GenBank. Nucleotide sequence of the fragment [GenBank Accession No. EU167936] showed more than 98% identity with other isolates [GenBank Accession Nos.:AJ748256;AJ564636;AB053256;AF531458;AJ564637 ; AJ748257]. It also showed more than 90% identity with other isolates from lily[AJ874695;AJ310203;EF158108;AB054886;AM048875;AJ874696; AJ879511; S60810; AY464944]Remarkably high Similarity in the nucleotide has been observed despite of their different origins. The virus is an important virus pathogens of crops which is harmful to lily and tulip, no reports on the Narcissus.2. Narcissus yellow stripe virus (NYSV) is a potato Y virus (Potyvirus) virus, which is widely considered to be pathogen-specific infection of narcissus. Its typical symptoms is yellow along the leaf veins, Thin plants in serious condition, dwarf, small bulb. In this study, NYSV was detected using RT-PCR technology to quickly and efficiently from Zhangzhou narcissus. It is not known of the reports about the rapid detection of Narcissus yellow stripe virus. In this study , the rapid and efficient detection of the virus on narcissus in Zhangzhou is very important for Narcissus virus disease control. It is also very important that this variety of resources for the protection of Narcissus, the achievement of sustainable development to provide technical support for prevention and treatment of disease.3. This test focused on a more simple and convenient,efficient and effective method that can greatly reduce the cost of production of virus-free.it also can be used to overcome the problem of low survival rate caused by the conventional "hot treatment + shoot tip culture" virus-free plants approach. LSV-free technology in the study, the use of commodity ball "big tip" high-temperature and variable-temperature (during the day and 38℃-10h, at night 32℃-14h) dark-wide training. A month later, virus-free rate was 100%. And salicylic acid for a certain degree of inhibition of virus, but failed to remove the virus, to be further studied.
